摘    要:目的采用反相高效液相色谱法测定阿霉素聚乳酸微球中的含量。方法采用AgilentEclipseXDB-C18色谱柱(150mm×4.6mm,5μm),以乙腈-甲醇-水(4∶5∶1)为流动相,检测波长为233nm。结果阿霉素在5~50μg/ml与峰面积呈良好的线性关系(r=0.9994),方法平均回收率为99.92%,RSD=0.82%。结论反相高效液相色谱法测定阿霉素聚乳酸微球的含量简便易行,专属性强,灵敏度高,重复性好,结果准确。

Abstract:Objective To determine the loading amount of doxorubicin polylactic acid microspheres by RP- HPLC. Methods An RP-HPLC method was employed for determination using Agilent Eclipse XBD-C18 column (150 mm×4.6 mm, 5 μm) with a mobile phase consisted of acetonitrile-methanol-water (4∶5∶1). The length of wavelength was 233 nm. Results Good linearity was achieved in the range of 5~50 μg/ml (r=0.9994). The recovery was 99.92% with RSD of 0.82%. Conclusion This method appeared easy-to-use, specific and sensitive, reproducible and accurate.
