丁基苯酞对慢性脑缺血大鼠学习记忆能力及前脑组织线粒体Na+-K+-ATP酶的影响

摘    要:目的 探讨丁基苯酞对慢性脑缺血大鼠学习记忆能力以及前脑组织线粒体Na+-K+-ATP酶的影响.方法 将实验用SD大鼠随机分为假手术组(n=10)、模型组(n=15)、丁基苯酞大剂量治疗组(n=15)和小剂量治疗组(n=15).采用双侧颈总动脉结扎制作慢性脑缺血模型,4周后,分别给模型组、大、小剂量丁基苯酞组大鼠腹腔注射生理盐水、丁基苯酞6 mg/kg和2 mg/kg,2次/d.用药4周后,利用Morris水迷宫试验测试大鼠学习记忆能力,提取前脑组织线粒体,测定线粒体Na+-K+-ATP酶的活性.结果 较假手术组相比,模型组大鼠记忆能力明显下降,逃避潜伏期延长分别为(10.41±3.81)s和(25.54±9.82)s,P<0.01]线粒体ATP酶活力下降分别为(3.73±0.32)μmol·mgprot-1·h-1和(2.81±0.40)μmol·mgprot-1·h-1,P<0.05],而丁基苯酞大、小剂量组较模型组记忆能力显著增强分别为(11.72±5.78)s和(12.48±5.45)s,P<0.01],ATP酶活力明显增加分别为(3.53±0.37)μmol·mgprot-1·h-1和(3.43±0.43)μmol·mgprot-1·h-1,P<0.05],不同剂量丁基苯酞组差异无显著性(P>0.05).结论 丁基苯酞注射液可增强缺血区神经元线粒体Na+-K+-ATP酶活力,改善线粒体能量代谢,提高慢性脑缺血大鼠的学习记忆能力.

Effects of dl-3-n-butylphthalide on learning-memory ability and Na + -K + -ATPase activity of mitochondria in rats with chronic cerebral hypoperfusion

Abstract:Objective To explore the effects of dl-3-n-butylphthalide on learning-memory ability and Na+-K+-ATPase activity of mitochondria in rats with chronic cerebral hypoperfusion.Methods Sprague-Dawlay rats were randomly divided into sham group( n= 10) ,model group (Permanent occlusion of bilateral common cawater maze test.Na+-K+-ATPase activity in mitochondria of forebrain tissue were also measured.Results Compared with sham group, memory ability of rats in model group decreased obviously( ( 10.41 ± 3.81 ), ( 25.54 prot- 1 , p <0.05 ).The memory ability of rats in large/small dose dl-3-n-butylphthalide groups both increased notably compared with the model group( respectively( 11.72 ± 5.78 ) s and ( 12.48 ± 5.45 ) s, P < 0.01 ) and Na + -0.05 ).The effectives of large dose dl-3-n-butylphthalide and that of small dose dl-3-n-butylphthalide have no significantly differences.Conclusion Dl-3-n-butylphthalide could increase the Na + -K +-ATPase activity and protect mitochondria, improve the learning-memory ability of rats with chronic cerebral hypoperfusion.
