血管生成素-2和缺氧诱导因子-1α在膀胱移行细胞癌中的表达及意义

摘    要:目的:初步探讨血管生成素-2(Ang-2)和缺氧诱导因子-1α(HIF-1α)在膀胱移行细胞癌中的表达及其与膀胱移行细胞癌病理分级、临床分期之间的关系。方法:选取经临床病理诊断的膀胱移行细胞癌标本64例,正常膀胱组织19例.应用免疫组织化学S-P法检测Ang-2和HIF-1α在膀胱移行细胞癌中的表达情况.并与临床资料相对照进行统计学分析。结果:膀胱移行细胞癌中Ang-2和HIF-1a的阳性率分别为46.9%(30/64)、42.2%(27/64),而正常膀胱组织均无表达。Ang-2阳性率随其病理分级和临床分期增加而升高,G1与G3相比,差异有统计学意义(P〈0.05);浅表型和漫润型相比,差异有统计学意义(P〈0.01)。HIF-1。阳性率亦随其病理分级和临床分期增加而升高,G1与G2相比,差异有统计学意义(P〈0.05);浅表型和浸润型相比,差异有统计学意义(P〈0.01)。Ang-2与HIF-1α的等级相关系数rs=0.498(P〈0.01)。结论:Ang-2和HIF-1α过表达均与膀胱移行细胞癌的恶性程度和浸润进展有关。膀胱移行细胞癌中Ang-2和HIF-1α过表达具有正相关关系;可能两者相互作用,通过影响肿瘤血管形成,共同参与膀胱移行细胞癌的发生、发展。

Abstract:Objective:The study is to investigate the relationships among the expression of Ango2, HIF-1α and the clinicopathological variables in patients with transitional cell carcinoma(TCC) of the urinary bladder, the correlation of Ang-2 and HIF-1α in transitional cell carcinoma of the urinary bladder. Methods:The expression of Ang- 2 and HIF-1α were assessed by immunohistochemistry in tissue sections from 64 patiens with transitional cell carcinoma of the urinary bladder and in 19 normal bladder samples, as to compare and analyze the clinical cases as well as statistically analyze. Results Tbere is no marked expression of Ang-2 and HIF-1α in normal bladder samples. The positive expression of Ang-2 and HIF-la was detected in TCC samples. The positive rate of Ang-2 and HIF- la is respectively 46. 9 % (30/64), 42. 2 %(27/64). In TCC, positive Ang-2 expression was significantly correlated with histological grade(P〈0. 05), clinical stage(P〈0. 01), positive HIF-1α expression was significantly correlated with histological grade(P〈0. 05), clinical stage(P〈0. 01). There is relatively correlation in the overexpression of Ang-2 and HIF-1α in transitional cell carcinoma of the urinary bladder each other(rs=0. 498, P〈0. 01). Conclusions: These results suggest that there is no expression of Ang-2 and HIF-1α in normal bladder tissues. The overexpression of Ang-2 and HIF-1α are associated with tumour grades, stages. There is positive correlation between the expression of Ang-2 and HIF-1α in TCC. These findings suggest that Ang-2 and HIF-1α all possibly play an important role in development and progression of TCC.
