愈障汤对慢性再生障碍性贫血患者骨髓CD_(34)~+细胞bcl-2表达的影响

目的　探讨愈障汤治疗慢性再生障碍性盆血 (CAA)的作用机制。方法　对愈障汤治疗的 39例CAA患者分别于治疗前后采用磁式细胞分选器 (MACS)分离纯化骨髓CD3 4+ 细胞 ;采用SABC亲和细胞法和CMIAS系列多功能真彩图像分析仪检测骨髓CD3 4+ 细胞bcl 2的表达 ,并与 2 6例健康对照组比较。结果　CAA患者骨髓CD3 4+ 细胞bcl 2蛋白表达的阳性率和OD值治疗前均较健康组减低 (P <0 .0 1) ,治疗后显著升高 (P <0 .0 1) ,bcl 2蛋白表达与临床疗效呈正相关。结论　愈障汤治疗CAA的作用机制可能是通过上调凋亡抑制基因bcl 2的表达 ,从而阻抑了骨髓CD3 4+ 细胞的过度凋亡。

Effect of Yuzhang Decoction upon bcl-2 Expression on CD_(34)~+ Bone Marrow Cells in Chronic Aplastic Anemia

Objective To investigate the mechanism of Yuzhang decoction(YZD) in treating chronic aplastic anemia(CAA).Methods CD 34 + bone marrow cells were separated and purified from 39 CAA patients before and after the treatment with YZD respectively.bcl 2 expression in CD 34 + bone marrow cells was assayed with SABC technique and CMIAS multi function color photo instrument.The results were compared with those of the normal control group.Results Before the treatment,the positive rate and OD number of CD 34 + bone marrow cells in CAA were lower than those in the normal control group(P<0.01),but increased significantly after the treatment(P<0.01).bcl 2 protein expression was positively correlated with the clinical effect.Conclusion YZD in treating CAA might increase bcl 2 protein expression that inhibits apoptosis,thus inhibit apoptosis of CD 34 + bone marrow cells.
