Objective To establish a high-throughput(HTS) screening model targeting inosine monophosphate dehydrogenase(IMPDH) for the discovery of novel antitubercular drugs.Methods The H37Rv IMPDH coding gene guaB2 was amplified and cloned into pBEV expression vector.The recombinant GuaB2 protein was expressed in Escherichia coli BL21(DE3)pLysS and itsactivity was measured at 340 nm wavelength absorbance.HTS screening model was established based on the activity of GuaB2 and Z' factor was used to evaluate the quality ...
