大鼠完全脊髓横断动物模型的建立及完整胚胎脊髓移植后功能恢复的研究

摘    要:
目的建立有效、可靠的完全脊髓横断动物模型,探讨多个完整胚胎脊髓移植联合脑源性神经营养因子(BDNF)对脊髓完全横断损伤的修复作用。方法将Wistar雌性大鼠分为正常对照组,手术组A组(单纯横断)、B组(横断+BDMF)、C组(横断+移植)、D组(横断+移植+BDNF)。手术显微镜直视下切除大鼠脊髓3-4mm,镜下观察确保完全离断,取3—4个完整胚胎脊髓同时移植。B组及D组应用微型泵定时定量泵入BDNF溶液(2mg/ml)。术后1—6周进行功能锻炼及行为学分析。结果术后A组、B组后肢运动功能未恢复;术后3W,C组及D组开始恢复后肢运动功能,术后6W出现协调踏步动作。术后1,2W,C组与D组CBS行为学评分无显著性差异(P〉0.05),术后3W、4W,CBS评分差异有显著性(P〈0.01)。结论本实验成功的建立了大鼠完全脊髓横断模型。多个完整胚胎脊髓组织移植解决了脊髓完全离断情况下所需移植物总量较大的问题,联合应用BDNF,对脊髓损伤修复效果更为理想。

Abstract:
Objective To establish effective and reliable complete spinal cord transverse animal models and approach effect of several integrated fetal spinal cord grafts associated with BDNF on repair of complete spinal cord transection in rats.Methods Divided Wistar female rats into normal control and operation groups which were divided into A group(transaction only),B group(transaction+BDNF),C group(transaction+transplantation),D group(transaction+transplantation+BDNF).We cut down a whole spinal cord segment of 3-4 mm with sharp corectome by looking straightly at the operating microscope,and ensured complete mutilation microscopically,took 3-4 integrated fetal spinal cord to be transplanted simultaneously.B group and D group were embedded with advanced Alzet osmotic pumps to inject BDNF solution by timing and quantitation(2 mg/ml).During the 1-6 week post-operation rats had done functional exercise and been evaluated on the aspect of raxiology.Results The motor function of A and B groups didn't recover at the post-operation.At the week 3-4 post-operation the hind limb motor function of C and D groups began to recover,at the week 6 post-operation the two groups showed concordant stepping action.At the week 1,2 post-operation CBS score of C group was not significant difference from the one of D group(P>0.05),but at the week 3,4 post-operation their CBS score was significant difference(P<0.01).Conclusion Our experiment established complete spinal cord transaction model of rats successfully.Several integrated fetal spinal cords solved the question that complete myelonic mutilation needed larger total amount of grafts.Effect of transplantation associated with BDNF was more ideal on repair of SCI.
