IL-10基因修饰树突状细胞对大鼠皮肤移植免疫耐受的诱导

摘    要:目的:探讨白介素10(IL-10)修饰的供体树突状细胞(dendriticcells,DC)对大鼠同种异体皮肤移植术后免疫耐受的诱导效果,为该基因治疗方法在皮肤移植的临床应用提供依据。方法:供体DC以humanIL-10基因修饰后,通过外周静脉输入受体体内,一周后行大鼠同种异体皮肤移植,根据实验分组观察受体移植皮肤存活情况。结果:对照组、未修饰DC组、hIL-10修饰DC组大鼠移植皮肤存活时间分别为(6.86±1.34)d、(7.57±2.45)d、(13.71±4.04)d,经统计学处理,未修饰DC组与对照组之间无显著性差异,而hIL-10修饰DC组与对照组之间有显著性差异(P<0.01)。结论:hIL-10修饰的供体DC预处理受体,可明显延长移植皮肤存活时间。

Abstract:Objective The effect of donor human IL-10 modified dendritic cells (DC) on allograft immune tolerance was explored to provide theoretical and practical basis for anti-rejection of skin transplantation. Methods The recipient was pretreated with donor human IL-10 modified DC or with unmodified DC, skin transplantation was performed and the survival time of skin allograft were observed. Results The survival time in the group of control,unmodified DC and hIL-10 modified DC were (6.86±1.34) d,(7.57±2.45) d,and (13.71±4.04) d respectively.Statistical analysis showed that there was no obvious difference between unmodified DC and control group. However, the survival time of skin allograft in hIL-10 modified DC group had significant difference compared with control (P<0.01). Conclusion The pretreated recipient with donor hIL-10 modified DC can prolong the survival time of rat skin allograft after transplantation.
