玉屏风散含药血清影响胸腺细胞凋亡的时效关系

摘    要:
目的:应用中药血清药理学方法,通过时间-效应关系研究,观察玉屏风散(YPFP)对地塞米松(DEX)诱导的小鼠胸腺细胞凋亡的影响。方法:9组小鼠按0.1 ml/10 g体重用玉屏风散水煎液灌胃,剂量相当于48.0 g饮片/kg体重,于给药后15 min,30 min,1 h,1.5 h,2 h,2.5 h,3 h,4 h分别在无菌条件下取血,分离含药血清。另设蒸馏水灌胃后的血清做空白对照组。取正常胸腺细胞加入地塞米松,再加入不同时相的含药血清和空白血清分9组培养,进行凋亡率检测,并绘出凋亡率时-效关系变化曲线。结果:玉屏风散在60~120 min时相含药血清具有明显抑制胸腺细胞凋亡的作用(P<0.01或P<0.05),60min时相含药血清的抑制作用最强,为达峰时相。结论:玉屏风散含药血清对胸腺细胞凋亡有一定免疫调节作用。

Abstract:
Objective: With a method of serum-pharmacology to study the effect of Yu-Ping-Feng powder (YPFP) on apoptosis of thymocytes induced by dexamethason (DEX) in mice. Methods: 9 groups of healthy mice were given YPFP apozem orally at a dose of equivalent to 48. 0 g of herbal slices per-kilogram weight. Blood were collected in an aseptic state to disaffiliate serum at 0. 25 , 0. 5 , 1, 1. 5, 2, 2.5, 3, and 4 hrs after the treatment. One more group was given distilled water instead, whose extracted serum was used as a control. The immunopharmacological effect of YPFP was determined with these sera. The thymocytes were incubated respectively for 3, 6, 9, 12, and 24 hrs with 1) DEX; 2) DEX plus YPFP; and 3) YPFP. The thymocytes apoptosis was studied under microscopes. Results: It was found that apoptosis of thymocytes could be induced by DEX. It was also found that the apoptosis induced by DEX was markedly inhibited by the drug-containing serum taken at 1 - 2 hrs after the treatment (P <0.01, P <0.05) , specially the serum of l hr, but the spontaneous apoptosis of thymocytes was not affected. Conclusions: The drug-containing serum of mice taken at the time of l ~ 2 hrs after ingestion of YPFP apozem could regulate the apoptosis of thymocytes.
