摘    要:目的 :探讨血液流动产生的切应力对白细胞 血管内皮细胞 (VEC)相互粘附所产生的影响及其在炎症、免疫反应、动脉粥样硬化发生发展中所起的重要作用。方法 :将已汇合的人脐静脉内皮细胞玻片放于平板流动腔中 ,以层流不同切应力 0 .6、1.2、2 .4Pa剪切不同时间 (2、8、12h) ,用免疫细胞化学方法检测VEC表面ICAM 1表达 ,以显微镜下计数阳性细胞率表示。结果 :稳层流切应力能上调HUVECICAM 1表达 ,切应力是内皮细胞粘附分子表达的调节因素之一。

Abstract:Objective: To study the effect of laminer shear stress on expression of intercellular adhesion molecular-1(ICAM-1) in vascular endothelial cells. Method: Human umbilical vein endothelial cells were subjected to controlled levels of laminer shear stress( 0.6, 1.2, 2.4 Pa) in a parallet plate flow chamber. Expresion of ICAM-1 were measured by immunochemistry. Results: Expression increased 125.4% or 142.03% when endothelial cells were presheared for 8 h at 1.2 or 2.4 Pa. The increased expression of ICAM-1 by shear stress was time and force dependent. Conclusion: The results showed biomechanical force in addition to humoral stimuli acte as a regulator of cell adhesion molecule expression in vascular endothelial cells.
