p53 codon72多态性与延边地区乳腺癌发生的相关性

[目的]探讨p53基因codon72多态性与延边地区朝鲜族及汉族乳腺癌发生的遗传易感性关系,并观察多态性分布是否存在种族差异.[方法]采用限制性酶切片段多态性分析技术检测90例乳腺癌组织及94例对照健康妇女p53 codon72基因多态性的分布.[结果]p53 codon72的3种基因型Arg/Arg、Pro/Pro、Pro/Arg在朝鲜族乳腺癌组中所占比例分别为48.78%,29.27%,21.95%,与朝鲜族对照组的16.28%,46.51%,37.20%相比较,总构成比差异有统计学意义(χ2=10.17),其中朝鲜族乳腺癌组Arg/Arg所占比例明显高于对照组,且高于其他2个型;在延边地区汉族乳腺癌组中所占比例分别为20.41%,32.65%,46.93%,与汉族对照组33.33%,43.13%,23.53%相比较差异有统计学意义(χ2=6.18),Pro/Arg在汉族乳腺癌组中所占比例明显高于对照组.[结论]p53 Arg/Arg及Pro/Arg基因型分别可能是延边地区朝鲜族及汉族乳腺癌发生的主要遗传易感因素.

Study on correlation between p53 codon 72 polymorphism and breast Cancer in Yanbian area

OBJECTIVE To study the correlation between p53 codon 72 polymorphism and genetic susceptibility for breast cancer in Korean-Chinese and Han nationality in Yanbian area,and to observe that the polymorphic distribution of p53 codon 72 among nations whether or not existed the difference among nationalities.METHODSThe distribution of p53 gene type in 90 block of paraffin embedded tissue of the breast cancer and 94 of venous blood of health women was detected by PCR-RFLP methods.RESULTSThe proportion of three types of Arg/Arg,Pro/Pro,Pro/Arg in p53 codon 72 gene were 48.78%,29.27%,21.95% in breast cancer group of Korean-Chinese,and 16.28%,46.51%,37.20% in control group,respectively,and total of constituent ratio was significant difference between two groups(χ2=10.17),in whitch the ratio of Arg/Arg was significantly higher in breast cancer group of Korean-Chinese than in control group and the other 2 types.The proportion of three types of Arg/Arg,Pro/Pro,Pro/Arg in p53 codon 72 gene were 20.41%,32.65% and 46.93% in breast cancer group of Han nationality,and 33.33%,43.13% and 23.53% in control group,respectively,and the difference was statistically significant between two groups(χ2=6.18),the ratio of Pro/Arg was significantly higher in breast cancer group of Han nationality than in control group.CONCLUSIONThe genotype of Arg/Arg and Pro/Arg in p53 codon 72 could be a genetic predisposing factor for development of breast cancer in Korean-Chinese and in Han nationality of Yanbian area,respectively.
