血清瘦素及瘦素受体基因Gln223Arg多态性与妊娠期糖尿病的相关性研究

目的:探讨血清瘦素水平及瘦素受体基因Gln223Arg多态性与妊娠期糖尿病的相关性。方法:应用放射免疫分析法(RIA)及聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)技术,检测62例妊娠期糖尿病(GDM)患者及60例正常孕妇(NGT)血清瘦素及瘦素受体基因Gln223Arg多态性。结果:(1)GDM组血清瘦素水平为14.2±4.3μg/L,NGT组为10.1±2.6μg/L,二者差异有统计学意义(P<0.05)。(2)GDM组孕妇瘦素受体基因Gln223Arg基因型GG、(GA+AA)频率分别为61.29%、38.71%,正常孕妇组分别为81.67%、18.33%,两组比较差异有统计学意义(P<0.05);GDM组孕妇瘦素受体基因Gln223Arg等位基因频率G、A分别为77.42%、22.58%,正常孕妇组孕妇分别为89.17%、10.83%,两组比较差异有统计学意义(P<0.05);(3)GDM组中基因型为GA+AA的孕妇瘦素水平为15.3±4.1μg/L,GG基因型为13.5±3.7μg/L,二者差异有统计学意义(P<0.05)。结论:瘦素,瘦素受体基因Gln223Arg多态性与GDM相关,GDM患者瘦素受体基因Gln223Arg变异可能影响血清瘦素水平。

Association of serum leptin and Gln223Arg polymorphism in leptin receptor gene with gestational diabetes mellitus

Objective:To investigate the association of serum leptin and Gln223Arg polymorphism in leptin receptor gene with gestational diabetes mellitus.Methods:The serum leptin level and Gln223Arg polymorphism in leptin receptor gene were determined by radio immunoassay and polymerase chain reaction-restriction fragment length polymorphisms(PCR-RFLP) assay in 62 patients with gestational diabetes mellitus(GDM) and also in 60 normal pregnant women.Results:(1)The serum leptin levels in pregnant women with GDM and normal pregnant women were 14.2±4.3μg/L and 10.1±2.6μg/L respectively,suggesting the serum leptin levels were significantly higher in pregnant women with GDM than that in normal controls(P<0.05).(2)The frequencies of GG,(GA+AA) genotypes of leptin receptor gene Gln223Arg were 61.29% and 38.71% in GDM group respectively,and they were 81.67% and 18.33% in normal group,respectively(P<0.05).The frequencies of two alleles G and A of leptin receptor gene Gln223Arg were 77.42% and 22.58% in GDM group,while 89.17% and 10.83% in normal group.The frequency of the allele A was higher in GDM group than that in normal group(P<0.05).(3)The serum leptin level was 15.3±4.1μg/L in GDM group with GA+AA genotypes.It was 13.5±3.7μg/L in GDM group with GG genotype(P<0.05).Conclusion:Leptin and Gln223Arg polymorphism in leptin receptor gene were associated with GDM.The Gln223Arg polymorphism in leptin receptor gene could affect the level of leptin in GDM group.
