血必净对哮喘小鼠气道重塑及黏附分子ICAM-1、VCAM-1 的影响

摘    要:
目的 探讨血必净注射液对哮喘小鼠气道重塑及肺泡灌洗液中黏附分子ICAM-1、VCAM-1 的影响。方法 40 只BALB/c 小鼠随机分成健康对照组(C 组)、慢性哮喘组(A 组)、血必净干预1 周组(1 组)、血必净干预2 周组(2 组)和血必净干预3 周组(3 组),每组8 只。复制哮喘模型后以血必净注射液分别干预1、2、3 组小鼠1、2 和3 周。麻醉处死后取肺泡灌洗液(BALF)及肺组织。BALF 以酶联免疫吸附实验(ELISA)法测定细胞间黏附分子1(ICAM-1)、血管内皮细胞黏附分子1(VCAM-1)的含量,肺组织切片并染色,镜下观察肺组织病理形态改变及气道形态学改变。结果 ①血必净干预后可降低哮喘小鼠BALF 中ICAM-1、VCAM-1 的水平,3 组与其他组比较,差异有统计学意义(P <0.05),降低最显著。②镜下观察见血必净可改善哮喘小鼠气道及肺组织的病理形态,3 组效果最明显。③ 1 组、2 组、3 组支气管壁厚度、平滑肌厚度逐渐变薄,但均厚于C 组,差异有统计学意义(P <0.05)。结论 血必净可改善哮喘小鼠气道重塑并可通过下调ICAM-1、VCAM-1 的水平改善气道炎症。

Abstract:
Objective To discuss the effect of Xuebijing injection on airway remodeling and theexpression of adhesion molecule content of both VCAM-1 and ICAM-1 in bronchoalveolar lavage fluid(BALF) of mice with asthma. Methods Forty BALB/c mice were divided into 5 groups, including acontrol group (group C), an asthma model group (group A), a Xuebijing intervention for 1 week group(group 1), a Xuebijing intervention for 2 weeks group (group 2), a Xuebijing intervention for 3 weeksgroup (group 3), with eight mice in each group. The asthma model was established, then the groups 1,2 and 3 accepted Xuebijing intervention respectively for 1, 2 and 3 weeks. The lung tissues and BALFwere got after the mice were sacrificed with anesthesia. The content of VCAM-1 and ICAM-1 in BALFwas detected by ELISA. Pathological changes of the lung tissues and morphological changes of airwaywere observed under light microscope after staining in lung slices. Results The mice in the groups 1, 2and 3 had lowered levels of VCAM-1 and ICAM-1, those in the group 3 decreased the most obviouslycompared to the groups 1 and 2 (P < 0.05). Xuebijing injection could improve the pathological changes of airway and the lung tissues in the mice with asthma, the group 3 had the most obvious effect. Thebronchial wall thickness and the smooth muscle wall thickness were gradually declined from the group1, group 2 to group 3, but all were thicker than those of the group C and thinner than those of the groupA (P < 0.05). Conclusions Xuebijing injection can improve airway remodeling, and improve airwayinflammation by influencing the levels of VCAM-1 and ICAM-1.
