宫颈鳞癌组织中c-fos 蛋白表达与HPV 感染的相关性

摘    要:目的 探讨宫颈鳞癌及宫颈病变组织中原癌基因c-fos 蛋白和人乳头状瘤病毒(HPV)表达及其与宫颈鳞癌发生、发展的关系。方法 选取承德医学院附属医院妇科门诊及病房经病理确诊为宫颈鳞癌和宫颈病变组织患者共148 例。依据病理学诊断结果分为慢性宫颈炎症组(31 例)、低级别鳞状上皮内病变(LSIL)组(35 例)、高级别鳞状上皮内病变(HSIL)组(33 例)及宫颈鳞状细胞癌组(49 例)。分别采用PCR- 反向点杂交技术及免疫组织化学PV 二步法检测各组织中HPV-DNA 和c-fos 蛋白表达。结果 宫颈鳞癌组织中HPV 感染和c-fos 蛋白表达高于慢性宫颈炎症组、LSIL 组及HSIL 组(P <0.05);宫颈鳞癌组c-fos 蛋白表达与肿瘤的分化程度、TNM 分期、有无淋巴结转移密切相关(P <0.05);HPV 阳性宫颈鳞癌患者c-fos 蛋白表达率高于HPV 阴性患者(P <0.05)。结论 c-fos 蛋白在宫颈鳞癌及宫颈病变组织均有一定程度表达,随宫颈病变程度的加重c-fos 蛋白表达逐渐升高,宫颈鳞癌组织中c-fos 表达与HPV 感染密切相关。

Abstract:Objective To explore the relationships of the expression of c-fos protein and Human Papillomavirus(HPV) in cervical squamous cell carcinoma and cervical lesions with the occurrence and development of cervicalsquamous cell carcinoma. Methods The study involved 148 patients who were diagnosed pathologically withcervical lesions or cervical squamous cell carcinomas in the Department of Gynecology of Affiliated Hospital atChengde Medical College. They were divided into a chronic cervicitis group (31 cases), a low-grade squamousintraepithelial lesion group (35 cases), a high-grade squamous intraepithelial lesion group (33 cases) and a cervicalsquamous cell carcinoma group (49 cases). PCR-reverse dot blot hybridization was used to detect HPV-DNA.Immunohistochemical PV method was adopted to detect the expression of c-fos in cervical tissues. Results Theexpressions of HPV and c-fos protein in the cervical squamous cell carcinoma group were significantly higherthan those in the chronic cervicitis group, the low-grade squamous intraepithelial lesion group and the high-gradesquamous intraepithelial lesion group (P < 0.05). The expression rate of c-fos protein in the cervical squamous cellcarcinoma group was closely related to the differential degree of tumor, TNM stage and lymph node metastasis(P < 0.05). The c-fos protein expression rate in the HPV-positive patients with cervical squamous carcinoma was higher than that in the HPV-negative patients (P < 0.05). Conclusions There is a certain degree of c-fos proteinexpression in cervical squamous cell carcinoma and cervical lesions. With the progress of cervical lesions, theexpression rate of c-fos protein increases. The expression of c-fos protein has a close relationship with HPV infectionin cervical squamous cell carcinoma tissues.
