大鼠心肌钙黏蛋白表达的增龄变化

目的探讨大鼠心肌细胞发育过程中钙黏蛋白(cadherin)的分布方式以及心肌细胞中cadherin表达的增龄变化规律。方法应用S-P免疫组化方法分别检测幼年、青年、中年和老年大鼠的心肌细胞钙黏蛋白cadherin的表达。结果细胞钙黏蛋白cadherin在大鼠的心房、心室、乳头肌、室间隔等处均有表达。幼年时期cadherin阳性颗粒大部分集中在心肌细胞端-端的闰盘处,青年、中年和老年大鼠cadherin典型的分布在细胞心肌末端的闰盘处。结论在大鼠心肌发育过程中,cadherin的表达存在增龄变化;以cadherin介导的细胞黏合对闰盘的形成是必备的,黏合连接稳定了细胞间的联系并且有利于心肌细胞收缩的同步化。

Aging changes of cadherin expression in rat myocardium

Objective To research the spatiotemporal patterning of cadherin expression during development of rat cardiomyocytes and age related changes of cadherin expression. Methods Used immunohistochemical method,myocardial cadherins distribution was investigated in intant (20～25 d),young (3～5 months) adult (6～10 months) and aged (13～15 months) rat heart. Results The expression of cadherin was located in myocardium of afrial and ventricule and septum interventriculare and papillary muscles. The cadherin immunolabeling distributed in the transverse teminals of the infant rat cardiomyocytes. From young to aged rat heart,the classic cadherin was located in transversely orientated intercalated disk. Conclusions The present paper demonstrated that the cadherin expression existsprogressive ge related changes. Adherens junction is one of the prerequisites for the formation ofintercalated disk in rat myocardium. The mechanical coupling provided by adherens junctions isessential for the stable cell cell contact,thereby synchronizing contractions of the heart.
