四逆汤对局部脑缺血大鼠的保护作用及其神经酰胺机制

摘    要:目的:研究四逆汤对大鼠局部脑缺血后脑组织神经酰胺含量的影响。方法:采用大鼠大脑中动脉局部阻塞模型(MCAO),观察四逆汤对脑缺血大鼠丙二醛(MDA)和神经酰胺含量、以及超氧化物歧化酶(SOD)活性的影响。结果:与正常组比较,缺血模型组MDA含量和脑组织神经酰胺含量均显著升高;与缺血模型组比较,四逆汤组能提高SOD的活性,减少MDA的产生(P〈0.05),降低神经酰胺含量(P〈0.001)。结论:四逆汤能对局部脑缺血大鼠产生保护作用,其可能机制是减轻氧化损伤,减少神经酰胺的生成量。

Abstract:Objective to investigate the influence of sini decoction (SND)on the concentration of ceramide in brain tissue following focal cerebral ischemia.Methods:Model of middle cerebral artery occlusion (MCAO)was performed in adult male Sprague-Dawley (SD)rats.SD rats were randomly divided into sham,model and SND groups.We measured the activity of SOD,and the concentration of MDA and ceramide.Results:The concentration of MDA in model group was markedly higher than that in sham group.SND enhanced the activity of SOD,and reduced the concentration of MDA(P<0.05).The concentration of ceramide in model group was the highest.The concentration of ceramide in SND group was lower than that in model group(P<0.001).Conclusion:The mechanism by which SND showed robust neuroprotection against cerebral ischemia may be that SND could enhance the ability to suppress oxidative damage caused by focal cerebral ischemia,and reduce the concentration of ceramide in brain tissue.
