载辛伐他汀硫酸钙对去卵巢大鼠胫骨骨缺损修复的影响

摘    要:
目的观察辛伐他汀与硫酸钙混合后对骨质疏松骨缺损修复的促进作用。方法选用健康3月龄雌性SD大鼠48只,切除双侧卵巢,低钙饮食3月,并将胫骨上端造成3 mm骨缺损,随机均分为3组,未植骨组直接缝合皮肤,硫酸钙组在骨缺损处植入硫酸钙,载辛伐他汀硫酸钙组植入载辛伐他汀硫酸钙,植入后4、8周取标本进行组织学观察骨痂形成情况,植入后8周行Micro-CT检测,于12周对标本进行生物力学分析。结果植入4、8周时,载辛伐他汀硫酸钙组骨愈合程度较硫酸钙组和未植骨组有显著性改善,8周时,Micro-CT检测骨缺损区内骨体积分数(BV/TV),组织骨密度(t BMD),骨小梁数量(Tb.N),载辛伐他汀硫酸钙组优于后两组,12周时生物力学检测,载辛伐他汀硫酸钙组失效强度和刚度优于硫酸钙组和未植骨组(P0.05)。结论载辛伐他汀硫酸钙对骨质疏松性骨缺损具有良好的成骨和骨诱导作用,可以较好地促进骨缺损的修复。

Abstract:
Objective Simvastatin can promote osteoblast and inhibit osteoclast activity, and promote the healing of fracture. The aim of this study was to observe the effect of the complex of simvastatin and calcium sulfate on the repair of osteoporotic bone defect. Methods 48 healthy 3 month old female SD rats, ovariectomized and on low calcium diet for three months, had 3mm bone defect created at the upper tibia, and were randomly divided into 3 groups. Non-bone graft group had the skin sutured directly. Calcium sulfate group had calcium sulfate implanted at the bone defect while simvastatin loaded calcium sulfate group had simvastatin loaded calcium sulfate implanted. Histological observation of bone formation was conducted at 4 and 8 weeks. Micro-CT examination was performed at 8 weeks and biomechanical analysis was performed at 12 weeks.Results The degree of healing was significantly improved in the simvastatin loaded calcium sulfate group than the non-bone graft group and the calcium sulfate group at 4 and 8 weeks by histological observation. Bone volume fraction (BV/TV), bone density (tBMD) and trabecular bone volume (Tb.N) detected by Micro-CT in the bone defect region were better in the simvastatin loaded calcium sulfate group than the other two groups at 8 weeks. Also, biomechanical test revealed that the strength and stiffness were higher in the simvastatin loaded group than the calcium sulfate group and the non-bone graft group (P < 0.05). Conclusion Our results suggest that simvastatin loaded calcium sulfate have good osteogenic and osteo-inductive effects on osteoporotic bone defects, and can promote the repair of bone defects.
