P-AKT、VEGF、MVD在非小细胞肺癌的表达及相关性分析

摘    要:
目的观察磷酸化蛋白激酶B(P-AKT)、血管内皮生长因子(VEGF)及微血管密度(MVD)在非小细胞肺癌(NSCLC)组织中的表达,并通过研究癌组织中P-AKT与血管生成的关系,初步探讨P-AKT在肺癌发生发展及转移中的机制,为肺癌的早期诊断提供可供参考的新方法。方法用免疫组化法对61例NSCLC及20例肺良性瘤样病变周围正常组织中P-AKT、MVD、VEGF的表达进行检测,观察肺癌及正常组织中P-AKT、MVD、VEGF的表达及与临床病理各因素相互关系,分析NSCLC中P-AKT、MVD、VEGF之间的相关性。结果NSCLC组织中P-AKT蛋白阳性表达44.3%(27/61),明显高于正常组织0%(0/20;P<0.05),其蛋白表达与肿瘤的分化程度明显相关。VEGF的表达与TNM分期、组织类型、淋巴结转移明显相关;P-AKT蛋白表达阳性组的MVD值为27.19±1.33高于P-AKT阴性组的23.83±3.06(P<0.05),并与VEGF的表达呈正相关(P<0.05)。结论NSCLC组织中P-AKT的表达明显增高,其过表达与VEGF相关,P-AKT对肺癌血管生成的调节可能与上调VEGF有关。

Expression of P-AKT, VEGF and MVD in NSCLC and their relationship
Abstract:
Objective To investigate the expression of protein kinase B (P-AKT), vascular endothelial growth factor (VEGF) and microvascular density (MVD) in non-small cell lung cancer and normal lung tissue and their correlations, to study the new diagnosis method of lung cancer through the relationship of P -AKT and angiogenesis. Method Expression of P -AKT, VEGF and MVD was detected by Streptavidin-Peroxidase (SP) method in 61 cases of NSCLC and 20 normal lung tissue respectively,the relationship between such expression and clinicopathological features of NSCLC was also studied. Result 1) P-AKT protein was expressed in 27 (44.3%) of 61 cases of NSCLC,significantly higher than that of normal tissue (0% ,0/20), (P <0.05). There was a significant correlation between P -AKT expression and differentiation in NSCLC; 2) the MVD of P-AKT positive group was higher than that of negative group (27.19±1.33 vs 23.83±3.06, P<0.05), and there was positive correlation between the expression of P- AKT and VEGF(P<0.05). Conclusion 1) The expression of P-AKT is higher in NSCLC than in normal tissue, they participate in the happen and progress of NSCLC and may be also served as a definite diagnosis marker; 2) there is positive correlation between the overexpression of P-AKT and angiogenesis in NSCLC, it may lead to angiogenesis of NSCLC according to enhancing the expression of VEGF.
