基于水凝胶的内皮细胞三维培养

摘    要:
目的研究可注射温度敏感性水凝胶的生物相容性,探讨其做为三维培养材料的可行性。方法将人脐静脉内皮细胞株ECV304细胞接种于水凝胶,体外共同培养后进行形态结构观察,用MTT法检测ECV304细胞增殖活性,AO/EB试剂盒观察细胞凋亡,NO试剂盒测量细胞分泌NO等功能。结果ECV304细胞在水凝胶中生长及功能均比较理想。AO/EB双重染色有少量黄色荧光,显示仅有少量细胞凋亡。细胞在水凝胶中培养后上清液体中分泌的NO浓度与二维培养组之间差异显著性无统计学意义(P=0.948)。结论可注射温度敏感性水凝胶生物相容性好,是一种良好的三维培养材料,可作为种子细胞的载体应用于组织工程中。

Abstract:
Objective To study the biocompatibility of a novel tbermosensitive injectable hydrogel and investigate the possibility of hydrogel as scaffold materials in tissue engineering. Methods ECV304 cells were cultured in hydrogels in vitro, cell morphology was investigated and function cell prolferation was observed with MIT method, cell apoptosis were measured using AO/EB kit , NO kit was used to measure NO secretion in culture medium. Results ECV304 cells were attached to and extended on hydrogel, cell viability were not affected. AO/EB double-staining had little yellow fluorescence, which implied small amount of apoptosis whens ceils were cultured in hydrogel. There was no statistical significance of NO concentration between hydrogel and control groups (P = 0.948 ). Conclusions The injectable thermosensitive hydrogel was of good biocompatibility, and could be used as a scaffold materials and the carriers of seed cells in tissue engineering.
