人、犬和小鼠骨髓基质细胞的细胞骨架观察

摘    要:本文观察了人、犬和小鼠体外培养骨髓基质细胞细胞骨架之异同.实验结果表明,应用Coomassie blue染色可清晰显示细胞骨架结构.人和犬的骨髓基质细胞以成纤维样细胞为多.胞体呈梭形,胞浆内有粗大的细胞骨架.其纤维丝走向与细胞长轴一致.小鼠的骨髓基质细胞由成纤维样细胞、类巨噬细胞和类网状细胞混杂组成.后两类细胞中缺乏明显的细胞骨架结构.

Abstract:The organizations of cytoskeletal network of human,canine and murine bone marrow stromal cells were compared in a liquid culture system.Using Coomassie blue stain,the microfilame ntbundles and the microtubular fibers were,found in the cytoplasm of fusiform cells.These fibers might radiate out toward the cell periphery.The distribution of cytoskeleton was closely correlated with the cell shape.These cells showed the morphological characteristics of fibroblasts.In cultured cells were also found two other types of bone marrow stromal cells without obvious organization of cytoskeletal network.One were irregular shaped cells with several.pseudopodia.Their number increased significantly when extraneous macrophage colony stimulating factor (M-CSF) was added to the culture medium.They had the features of macrophages.Another type of stromal cells were large and mostly round in shape,they showed some morphological characteristics of reticular cells.The human and canine bone marrow stromal cells were primarily composed of fibroblastoid cells,while the murine ones were heterogeneous,the macrophage-like cells were rather populous.
