摘    要:目的 观察低氧条件下血小板衍生性生长因子 (PDGF)对于培养人脐静脉内皮细胞 (HUVEC)增殖的影响。方法 流式细胞仪分析细胞周期各时相分布 ,氚 -亮氨酸 (3 H -Leu)掺入实验评价蛋白质合成 ,免疫组织化学方法检测增殖细胞核抗原 (PCNA)水平。结果 低氧可显著减少S期及G2 /M期细胞数 ,使停留于G0 /G1 期HUVEC细胞数增加 ,降低HUVEC3 H -Leu的掺入量及PCHA水平。低浓度的PDGF(5ng/ml)对低氧条件培养的HUVEC细胞周期时相分布、3 H -Leu的掺入量及PCNA水平影响不明显 ;高浓度PDGF(1 0ng、2 0ng)加速HUVEC由G1 期S期的转换 ,显著增加低氧培养HUVEC3 H -Leu掺入量 ,PCNA水平 ,此作用呈剂量依赖性。结论 PDGF能够拮抗低氧所致的培养HUVEC增生能力的下降 ,但低浓度PDGF(5ng/ml)作用不显著 ,剂量越大 ,拮抗作用愈显著。

Abstract:Objective To study the effect of platelet derived growth factors on the proliferation of hypoxia culture endothelial cell. Methods The distribution of cell cycle in different time phases was analyzed by flow cytometry, protein synthesis was assessed by 3H-Leucine corporation, and proliferation cell nuclear antigen (PCNA) was tested by immunohistochemistry. Results Hypoxia reduced the amount of S phase and G 2/M phase, increased the amount of HUVEC at the G 0/G 1 phase, and decreased the 3H-Leucine corporation as well as the PCNA level of HUVEC. PDGF with low concentration (5ng/ml) had no obvious effect on the distribution of cell cycle in time phases, the amount of 3H-Leucine corporation, or the PCNA level. PDGF with high concentration could accelerate HUVEC transform from G 1 phase to S phase, and remarkably increase the 3H-Leucine corporation amount as well as the PVNA level of HUVEC under hypoxia conditions. PDGF showed dose dependence. Conclusion PDGF can inhibit the decrease of HUVEC proliferation, but it shows no obvious effect under low concentration (5ng/ml), and the more the dosage is, the greater the inhibiting power.
