人喉鳞癌组织中HPV16E7、CyclinD1和Rb蛋白的表达

目的:探讨喉鳞癌组织中人乳头瘤病毒(HPV)16型E7、CyclinD1和Rb蛋白表达的变化.方法:采用免疫组织化学SABC法检测80例喉鳞癌、30例声带息肉组织中HPV16E7、CyclinD1和Rb蛋白的表达状况.结果:声带息肉组织中HPV16E7未见表达,喉鳞癌组织中的表达率为34/80(P＜0.000 1);声带息肉与喉鳞癌组织中Cy-clinD1的表达率分别为2/30和42/80(P＜0.000 1),Rb的表达率分别为25/30和42/80(P＜0.05).HPV16E7与CyclinD1的表达与临床分期、淋巴结转移情况和5 a生存率有关(P＜0.05或0.01).喉鳞癌组织中HPV16E7与Rb的表达相关(P＜0.01).结论:HPV16感染和Rb、CyclinD1的异常表达与喉鳞癌发生、发展有关.HPV16E7与CyclinD1可作为判定喉鳞癌预后的指标.

Expression of HPV16E7, CyclinD1, and Rb in human laryngeal squamous cell carcinoma tissue

Aim: To study the relationship of human papilloma virus (HPV)16E7,CyclinD1, and Rb with carcinogenesis of human laryngeal squamous cell carcinoma(LSCC). Methods: Immunohistochemical SABC technique was used to examine the expression of HPV16E7,CyclinD1,and Rb in 80 cases of LSCC and 30 cases of polyp of vocal cord tissue.Results: In carcinoma group, the positive rate of HPV16E7 was 34/80, CyclinD1 was 42/80 and Rb was 42/80;in polyp of vocal cord group, the positive rate of CyclinD1 was 2/30, Rb was 25/30 and HPV16E7 was negatively expressed (P< 0.05). The expression of HPV16E7 and CyclinD1 were associated with clinical stage, lymph node metastases, and 5-year survival rate(P<0.05 or 0.01) in LSCC tissue.HPV16E7 expression in LSCC tissue was correlated with Rb expression ( P< 0.01). Conclusion: Infection of high risk HPV and the abnormal expression of CyclinD1 and Rb are closely related to the carcinogenesis and development of LSCC. The expression of HPV16E7 and CyclinD1 could be used to estimate the prognosis of LSCC.
