Abstract: | Objective To observe the effect of Shengxian decoction on heart rate,electrophysiology of atrionector and histopathology in rabbit with sick sinus syndrome,and explore its mechanism.Methods 30 healthy Japanese white rabbits were randomly divided into six groups,control group,model group,Xinbao pill group,high-dose Shengxian decoction group,middle-dose Shengxian decoction group and low-dose Shengxian decoction group,5 rabbits in each group.Control group and model group received distilled water 15 mL / d.Xinbao pill group received Xinbao pill 0.18g/(kg·d).High-dose Shengxian decoction group,middle-dose Shengxian decoction group and low-dose Shengxian decoction group received Shengxian decoction,86.2,43.1,21.5 g/(kg·d),respectively(Chinese medicine granules was dissolved in 15 mL with distilled water).After 7 days of continuous gavage,rabbits model of sick sinus syndrome in model group,Xinbao pill group,high-dose Shengxian decoction group,middle-dose Shengxian decoction group and low-dose Shengxian decoction group were established by 20% formaldehyde wet sinus node area.Heart rates(HR) in each group were observed before modeling and after the success of modeling 5,30 min,1hr and 2hr.SACT,SNRT and CSNRT in each group were measured.Pathological changes of sinus were observed.Results HR after modeling 5,30 min,1 and 2hr in model group were decreased in comparison with those before modeling and control group(P0.05).HR Xinbao pill group,high-dose Shengxian decoction group,middle-dose Shengxian decoction group and low-dose Shengxian decoction group were higher than that in model group(P0.05).HR in middle-dose Shengxian decoction group and low-dose Shengxian decoction group were lower than those in Xinbao pill group and high-dose Shengxian decoction group(P0.05).SACT,SNRT and CSNRT in model group were significantly prolonged compared with those of control group(P0.05).SACT,SNRT and CSNRT in Xinbao pill group,high-dose Shengxian decoction group,middle-dose Shengxian decoction group and low-dose Shengxian decoction group were significantly shorted compared with those of model group(P0.05).There were obvious differences between middle-dose Shengxian decoction group and low-dose Shengxian decoction group,Xinbao pill group and high-dose Shengxian decoction group on SACT,SNRT and CSNRT(P0.05).The changes of pathological sinus node under high magnification(×400) showed that damages of sinus node cells in each dose group were lighter than that in the model group.Conclusion Shengxian decoction can effectively fight against acute injury due to sick sinus syndrome,and protect atrionector.The mechanism may be through the dilation of blood vessels,improve blood supply of atrionector,and promote recovery of sinus node function;improve cell membrane ion channels and thus increase the automaticity of the sinus node. |
