高效液相色谱法测定苦参中红车轴草苷的含量

摘    要:
目的建立测定苦参中红车轴草苷含量的高效液相色谱方法。方法HYPERSIL BDS C18色谱柱(4.6mm×200mm,5μm),流动相为甲醇-0.2%磷酸水溶液梯度洗脱(MeOH%:0 min10%,20min40%),流速为1.0mL·min^-1,检测波长为310nm,柱温为25℃。结果红车轴草苷在0.446~8.920/μg线性良好,相关系数r为0.9999,平均回收率为97.94%,RSD<1%(n=6)。结论本方法操作简便,分离效果好,灵敏度高,重现性好,为控制苦参药材质量提供参考。

Abstract:
Objective To established an HPLC method for the determination of trifolirhizin in Sophora flavescens Air. Methods A column of HYPERSIL BDS C18 (4.6 mm× 200 mm, 5/μm) was used. The mobile phase was composed of methanol and 0.2% phosphoric acid with a linear gradient elution from 10%to 40%methanol in 20 minutes. The flow rate was 1.0 mL·min^-1 , and the detection wavelength was 310 nm. Results The calibration curve of trifolirhizin was in good linearity over the range of 0. 446~8. 920 μg , (r=0. 999 9). The average recovery was 97.94%, and RSD was less than 1%(n=6). Conclusion The method is simple, sensitive, accurate, reproducible, and can be used to control the quality of Sophora flavescens Air.
