丰白散对COPD模型大鼠肺组织中MMP-9及TIMP-1表达的影响

摘    要:目的观察丰白散对慢性阻塞性肺疾病(COPD)模型大鼠肺组织中基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶组织抑制剂-1(TIMP-1)基因表达的影响,探讨其治疗COPD的作用机制。方法取10只大鼠为正常组,不予任何处理,另取大鼠36只采用烟熏加气管内滴加脂多糖(LPS)方法复制COPD模型,取造模成功的30只大鼠随机分为模型组、沐舒坦组和丰白散组各10只,予相应干预28天后,用苏木素—伊红(HE)染色评估各组大鼠肺组织病理改变,逆转录聚合酶链式反应(RT-PCR)检测MMP-9、TIMP-1 mRNA表达水平。结果正常组肺泡结构完整,肺泡间隔无缺损;模型组肺泡结构破坏、融合、弹力纤维网破坏;沐舒坦组、丰白散组病理改变程度明显减轻。模型组大鼠肺组织MMP-9、TIMP-1 mRNA表达水平明显高于正常组(P<0.05);沐舒坦组、丰白散组大鼠肺组织MMP-9、TIMP-1 mRNA表达水平较模型组下降(P<0.05),2组间差异无统计学意义(P>0.05)。结论 MMP-9、TIMP-1过度表达,MMP-9/TIMP-1失衡在COPD气道重构中发挥重要作用;丰白散能下调MMP-9、TIMP-1 mRNA表达,调节MMP-9/TIMP-1失衡,干预气道重构。
