甲状旁腺组织中细胞周期素D1过度表达与Rb肿瘤抑制基因的表达和临床意义

摘    要:目的探讨甲状旁腺组织中细胞周期素D1(cyclin-D1)及视网膜母细胞瘤易感基因(Rb)的表达及临床意义。方法应用免疫组化技术对36例甲状旁腺腺瘤,10例正常甲状旁腺组织及10例甲状旁腺增生中cyclin-D1及Rb肿瘤抑制基因的表达进行检测。结果腺瘤组cyclin-D1的表达明显高于正常组及增生组(P<0.05),正常组同增生组比较差异无显著性(P>0.05)。Rb的表达正常组、增生组和腺瘤组三者之间比较差异均无显著性(P>0.05)。cyclin-D1过度表达和Rb表达与肿瘤大小、血清钙水平及甲状旁腺素(PTH)水平无关。结论cyclin-D1的过度表达在甲状旁腺腺瘤发生中起重要作用。cyclin-D1过度表达导致Rb磷酸化或表达下调而失去细胞抑制效应可能为甲状旁腺腺瘤发生的机制之一。

Abstract:Objective To investigate the overexpression of cyclin-D1 and the expression of Rb supressoroncogene in parathyroid tissues and the clinical significance.Methods The overexpression of cyclin-D1 and the expression of Rb in 36 parathyroid adenoma tissues and 10 proliferative tissues and 10 normal tissues was detected by ABC immunohistochemistry.Results The overexpression of cyclin-D1 was significantly higher in parathyroid adenoma group than in normal parathyroid group and proliferation group(P<0.05),but there was no significant difference between normal parathyroid group and proliferation group(P>0.05).The expression of Rb and the overexpression of cyclin-D1 was not related to the clinical data.Conclusion The overexpression of cyclin-D1 plays an important role in parathyroid adenoma.The loss of the ability of inhibiting the cell growth resulting from the decrease or phosphorylation of Rb induced by the overexpression of cyclin-D1 might contribute to the etiology of parathyroid adenoma.
