嗅鞘细胞移植对脊髓半横断损伤大鼠后肢运动功能的影响

摘    要:目的观察嗅鞘细胞移植物对脊髓半横断损伤大鼠后肢运动功能的影响。方法30只雌性成年SD大鼠制成脊髓半横断损伤模型,并随机分成5组立即进行移植,A组移植纯化的嗅鞘细胞(olfactory ensheathing cells,OECs)悬液,B组移植纯化的OECs培养上清液,C组移植未纯化的OECs,D组移植未纯化的OECs培养上清液,E组移植DMEM/F12培养液,术后6 w内进行BBB评分和IP实验,用单因素方差分析进行统计学分析。结果A组的BBB评分在3w后较其它组高,差异有统计学意义(P<0.05),A组的IP实验结果在4 w后较其它组高,差异有统计学意义(P<0.05)。结论纯化培养嗅鞘细胞移植能促进脊髓半横断损伤大鼠后肢运动功能恢复。

Abstract:Objective To observe the effect of different graft on hindlimb movement in rats with hemisection of spinal cord injury.Methods A total of 30 female adult healthy Sprague-Dawley rats were acutely transplanted after hemisection of spinal cord injury,randomly divided into five groups,group A was transplanted with suspension of purified OECs(olfactory ensheathing cells),group B was transplanted with medium from cultured purified OECs,group C was transplanted with suspension of impurified OECs,group D was transplanted with medium from cultured impurified OECs,group E was transplanted with DMEM/F12 culture fluid.Basso-Beattie-Bresnahan Locomotor Rating Scale and Inclined Plan Experiment were tested in 6 postoperative weeks.The One-way ANOVA was used to test intergroup differences.Results The BBB locomotor rating scale of group A was significantly increased than others after 3th postoperative week(P<0.05).The score of IP experiment of group A was significantly increased than others after 4th postoperative week(P<0.05).Conclusion The hindlimb movement functional recovery was promoted in rats with hemisection of spinal cord injury after transplantation of purified OECs.
