摘 要: | 目的 研究芎芍胶囊含药血清对人脐静脉内皮细胞(HUVEC)的增殖作用。方法 采用血清药理学和体外培养HUVEC的方法,通过倒置显微镜观察、MTT法和流式细胞术(FCM)检测芎芍胶囊高、中、低3个浓度含药血清对HUVEC增殖的影响。结果 MTT法检测结果显示高、中、低剂量的芎芍胶囊含药血清均有促进HUVEC增殖的作用,增殖率分别为54.14%、54.98%、33.77%;FCM法检测发现高、中、低剂量芎芍胶囊含药血清均可增加S期细胞比例,分别为52.14%、42.65%、37.08%;显微镜观察结果也显示各含药血清具有促进HUVEC增殖的作用。结论 芎芍胶囊能促进HUVEC的增殖,其促进细胞增殖的作用与浓度有一定的关系;提示芎芍胶囊的促血管新生作用可能是通过促进血管内皮细胞的增殖。

Abstract: | Objective To investigate the proliferation efficiency of Xiongshao capsule pharmaco - serum on human umbilical vein endothelial cells (HUVEC). Methods HUVECs were incubated in vitro. The Xiongshao capsule pharmaco - serum was identified to stimulate the proliferation of HUVEC by inverted microscope, MTT and flow cytometry (FCM). Results The high,medium and low doses of Xiongshao capsules pharmaeo - serum were found to promote the role of HUVEC proliferation,and proliferation rate were 54.14 %, 54.98% and 33.77%, respectively. Proportion of cells in S phase in high,medium and low dose Xiongshao capsules pharmaeoserum were increased by FCM, and they were 52. 14%,42. 65% and 37. 08% respectively. The promoting proliferation of HUVEC was found by microscopy. Conclusion Xiongshao capsule could promote the proliferation of HUVEC by promotion of vascular endothelial cell proliferation. |
