摘    要:目的探讨碱性成纤维生长因子对大鼠骨髓间充质干细胞分化为神经元的作用及信号通路.方法利用bFGF与不同生长因子和化学诱导剂组合诱导间充质干细胞(mesenchymal stem cells, MSCs)向神经元转化,观察分化过程中细胞形态、数目的变化,观察免疫细胞化学检测分化后细胞神经细胞标志蛋白和部分神经功能相关蛋白以及FGFR的表达情况,并计算转化率.结果①3组不同条件诱导后,MSCs均能分化为具有典型神经元形态的细胞,并明显表达NSE、MAP2和5-HT1D,但不表达GFAP;②bFGF预诱导组的转化率明显高于未预诱导组;③诱导后的神经元样细胞FGFR3表达明显增强.结论以DMSO和BHA为基础诱导剂,bFGF预处理可明显提高MSCs跨胚层分化为神经元样细胞的转化率,浓度为10 ng/ml的bFGF主要是通过FGFR3作用于MSCs.

Abstract:Objective To study the effects and signal pathway of basic fibroblast growth factor (bFGF) on mesenchymal stem cell (MSCs) differentiating into neuron-like cells. Methods MSCs were isolated from adult rat bone marrow, and expanded on the flask undifferentiated state for 6 passages. Different induction conditions were employed. Immunocytochemistry was used to detect the neuronal phenotype and the expressions of fibroblast growth factor receptor 1 (FGFR1) and FGFR3 in pre- and post-differentiated cells. The transdifferentiation percentage was calculated. Results Under different induction conditions, MSCs resumed typical neuronal morphology. There was a higher transdifferentiation percentage in the group pretreated by bFGF than that of the control. The expression of FGFR3, not FGFR1, increased during differentiation. Conclusion bFGF can promote MSCs differentiating into neuron-like cells in vitro. This signal transduction is mainly mediated by FGFR3 pathway.
