ADAMTS-1对博莱霉素所致肺成纤维细胞纤维化的影响

摘    要:
目的:探讨Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶(ADAMTS-1)对博莱霉素诱导的大鼠肺成纤维细胞纤维化的影响及其机制。方法实验分为六组,博莱霉素组(Ⅱ组)在大鼠肺成纤维细胞中加入博莱霉素建立肺纤维化模型,在加入博莱霉素的同时分别加1、2、4μg ADAMTS-1(Ⅲ、Ⅳ、Ⅴ组),并与只加4μg ADAMTS-1(Ⅵ组)和正常大鼠肺成纤维细胞(Ⅰ组)进行对照。加入博莱霉素及ADAMTS-1处理24 h后,运用实时定量聚合酶链反应(RT-PCR)检测结缔组织生长因子(CTGF)、Ⅰ型胶原(COLⅠ)mRNA表达量,酶联免疫吸附试验(ELISA)检测细胞培养上清液中CTGF、COLⅠ质量浓度。结果Ⅱ组CTGF、COLⅠmRNA表达量及细胞培养上清液中质量浓度明显高于其他各组,而Ⅲ~Ⅵ组与Ⅱ组比较,不但CTGF、COLⅠmRNA表达降低,而且细胞培养上清液中质量浓度也明显下降,以Ⅵ组最低,差异有统计学意义(P<0.05)。结论 ADAMTS-1在一定程度上呈剂量依赖性抑制博莱霉素诱导的大鼠成纤维细胞纤维化。

Abstract:
Objective To investigate the effect and mechanism of a disintegrin-like and metalloproteinase with thrombospondin typeⅠmotif(ADAMTS-1) on bleomycin-induced pulmonary fibrosis. Methods The experiment was divided into 6 groups. The bleomycin group (the groupⅡ) was added with bleomycin in rat lung fibroblasts to establish model of pulmonary fibrosis,meanwhile,1,2,4μg of ADAMTS-A(groupⅢ,groupⅣ,groupⅤ) was added respectively on the basis of bleomycin ,and compared with the group of 4μg ADAMTS-A(groupⅥ) and normal rat lung fibroblast(groupⅠ). After 24 h treatment of bleomycin and ADAMTS-1,real-time quantitative polymerase chain reaction(RT-PCR)was used to detect the expression quantity of mRNA in connective tissue growth factor(CTGF) and typeⅠcollagen(COLⅠ);while the enzyme linked immunosorbent assay(ELISA) was used to determine the concentration of CTGF and COLⅠin nutrient solution. Results The mRNA expression levels of CTGF and COLⅠand protein concentration of supernatant in cell culture medium in group Ⅱwere obvious higher than those in other groups,but the group Ⅲ,group Ⅳ,group Ⅴ and group Ⅵ were lower than the group Ⅱ not only on the mRNA expression of CTGF and COLⅠbut also on the protein concentration of supernatant in cell culture medium ,and the group Ⅵwas the lowest with statistically significant difference(P<0.05). Conclusion The ADAMTS-1 can inhibit the bleomycin-induced pulmonary fibrosis to a certain degree with a dose-dependent manner.
