摘    要:目的:建立以反相高效液相色谱(RP-HPLC)法测定猫豆中猫豆胍含量的方法。方法:色谱柱为PhenomenexTMC18(250mm×4.6mm,5μm),流动相为0.1mol·L-1冰醋酸-甲醇(95∶5),流速为1.0mL·mim-1,检测波长为324nm。结果:猫豆胍质量浓度在0.00590~0.0944mg·mL-1范围内与峰面积积分值呈良好的线性关系(r=0.9999);平均回收率为99.92%,RSD=1.68%(n=9)。结论:本方法简便、准确、专属性强、重复性好,可作为猫豆药材或含猫豆胍的药物制剂中猫豆胍的含量测定方法。

Abstract:OBJECTIVE: To establish a RP-HPLC method for the determination of mucunaguanide in Stizolobium cochinchinensis. METHODS: PhenomenexTM C18 column (250 mm×4.6 mm, 5 um) was used.The mobile phase consisted of 0.1 mol.L^-1 glacial acetic acid-methanol (95:5) at a flow rate of 1.0 mL.min^-1,and the detection wavelength was set at 324 nm. RESULTS: The linear range of mucunaguanide were 0.005 90-0.094 4 mg.mL^-1 (r=0.999 9)with average recovery of 99.92% (RSD= 1.68%, n= 9). CONCLUSION: The method is simple, accurate and specificity with good reproducibility. It can be applied for the content determination of mucunaguanide in S. cochinchinensis or pharmaceutical preparations contained mucunaguanide.
