短小棒状杆菌纯化物对小鼠骨髓瘤细胞的作用

摘    要:
目的初步探求短小棒状杆菌(CP)纯化物是否具有抗肿瘤作用及其免疫机理。方法短小棒状杆菌纯化物(酶解法)及短小棒状杆菌分别注入负瘤小鼠。氚标记胸腺嘧啶掺入法(3H-TdR)测小鼠脾淋巴细胞转化试验;流式细胞仪对T淋巴细胞亚群进行检测;mRNA表达法检测细胞因子TNF和INF-γ的活性。结果短小棒状杆菌纯化物(酶解法)及CP均可抑制肿瘤细胞生长。免疫功能显示,CP和酶解法纯化的CP都能刺激机体T淋巴细胞的转化(P<0.05)、CD3+、CD4+、CD8+数值显著增加及细胞因子的表达增强(P<0.05),且酶解法纯化的CP还能刺激机体CD8+值显著增加(P<0.05)。结论酶解法纯化短小棒状杆菌不仅抑制骨髓瘤细胞生长,并能显著的激活巨噬细胞系统,刺激机体的细胞免疫功能。

Abstract:
Objective To study the tumor suppression and the immune enhancement effect of perified corynebacterium parvum(CP) in the myeloma cell of mice and the probable mechanism.Methods The antitumor effect of the CP and CP perification on the myeloma cell was determined in the BABL/C mice;spleen lymphocyte cells transformation was tested by ~3H-TdR methods.The number of T clusters was analyzed by the flow cytometer.And cellular immune response was evaluated by mRNA of TNF and INF-γ.Results CP and the purification of CP could inhibit the growth of tumor cells,decrease the tumor weight and increase the spleen weight(P<0.05);increase the number of CD3~+,CD4~+,CD8~+ and the ratio of CD4~+ and CD8~+ cells in the spleen(P<0.05);But could not increase the number of NK cells.Conclusion The CP and the purification of CP could significantly inhibit the growth of tumor in vivo,and enhancing the immunological function may be one of the reasons for the anti-tumor mechanism.
