摘    要:
目的 利用Wistar大鼠烟雾吸入模型,观察被动吸烟对Wistar大鼠雌激素(E2)、孕激素(P4)水平及卵巢雌激素受体α(ERα、孕激素受体(PR)蛋白及mRNA的影响,为提倡生育期妇女避免被动吸烟提供新的理论依据.方法 32只健康雌性Wistar大鼠,随机分为对照组和实验组,每组16只.实验组吸烟3个月,对照组正常饲养.3个月后处死两组大鼠.酶联免疫吸附试验方法(ELISA)检测大鼠血清中E2,P4的水平;免疫组织化学及免疫印迹法检测大鼠卵巢中ERα,PR蛋白的表达,RT-PCR检测大鼠卵巢中ERα,PRmRNA的表达.结果 ELISA结果显示实验组大鼠血清E2,P4水平显著低于对照组;实验组大鼠卵巢组织中ERα表达明显低于对照组.PR主要表达在大鼠卵巢组织卵泡颗粒细胞的细胞质和细胞核中,呈棕黄色颗粒状分布,对照组大鼠卵巢组织中PR的表达明显低于对照组.实验组卵巢组织中ERα及PR蛋白表达明显低于对照组.实验组卵巢组织中ERα及PR mRNA的表达也低于对照组.结论 被动吸烟可引起大鼠血清中E2及P4水平下降,卵巢中ERα及PR的表达减少,提示被动吸烟可破坏卵巢的功能,引起卵巢生殖内分泌失调.

Abstract:
Objective Utilizing the model of Wistar rat Smoke inhalation model,and aimed to ohserve the effects of passive smoking on the level of estrogen and progesterone,protein expressions of ERα and PR and mRNA of Wistar rat ovaries, h can provide us with theorial evidence promoting of reproductive age women to avoid passive smoking. Methods Thirty two heahhy female Wistar rats were divided into the control group and the passive smoking group at random. the passive smoking group smoked for 3 months and the control group fed normally. Aider 3 months, put two groups of rats to death. The levels of E2 and P4 in serum were detected by the enzyme linked immunosorbent assay( ELISA). The expressions of ERα and PR protein were detected by immunohistochemistry and Weslern blot. the mRNA expression of ERα and PR were detected by RT-PCR. Results The enzyme, linked immuno-sorbent assay showed thai the levels of E, and Pa were obvious ly lower in serum of the passive smoking group than that of the control group. ERα expressions were significantly lower in the ovary of the passive smoking group than that of eontrol group. PR posilive material was mainly distributed in the cytoplasm and cell nucleus of lhe rat ovarian granulosa cells. PR expression was significantly lower in the ovary of the passive smoking group than that of control group. Expressions of ERα and PR protein in ovary of passive smoking group were significantly lower Ihan that of control group. Expressions of ERα and PR mRNA in ovary of passive smoking group were significantly lower than that of control group. Conclusion Passive smoking can degrade the levels of E2 and P4 in rat serum,decrease the expressions of ERα and PR protein and mRNA in rat ovary. The results showed passive smoking can damage ovarian rum.lion and cause disorders of reproduetive endocrine.
