T-2毒素对软骨细胞CD44表达的影响

摘    要:目的 研究T-2毒素对软骨细胞CD44表达的影响,为阐明T-2毒素损伤软骨组织的机制提供科学依据。方法 利用RT-PCR方法检测CD44 mRNA在软骨细胞中的转录情况;利用免疫组化方法检测软骨细胞表面CD44的表达情况。结果 在T-2毒素作用5d后,毒素组、毒素加硒组CD44 mRNA表达水平均低于对照和对照加硒组。同时免疫组化结果显示,毒素组体外再建软骨组织CD44表达最低,其次为毒素加硒组,对照加硒组最高。结论 T-2毒素对于软骨细胞的CD44表达具有明显的抑制作用。

Abstract:Objective CD44 is a kind of membrane protein expressing on chondrocyte, and has an important effect in cartilage matrix metabolism. T-2 toxin is a questionable virulence factor of KBD, a kind of endemic disease in China. Investigating the effect of T-2 toxin on CD44 expression of cartilage chondrocyte will be valuable to clarify the pathogenesis of KBD. Methods CD44 mRNA expression was measured by RT-PCR and immunohistochemistry were used to investigate CD44 expression on chondrocyte. Results Chondrocyte cultured without T-2 toxin expressed more CD44 mRNA than that of chondrocyte cultured with the toxin; Chondrocyte cultured with selenium expressed more CD44 mRNA than that of chondrocyte cultured without selenium. The ration between CD44 mRNA and GAPDH mRNA in control group was 3.337, but the ration in T-2 group was 1.392; The ration in control group with Se was 4.010, but the ration in T-2 toxin with Se was 1.570. The positive immunohistochemistry activity can be detected in all groups, but CD44 expression in control group is higher than that of T-2 group. Conclusions The results indicate that T-2 toxin can significantly restrain the cartilage chondrocyte CD44 expression. Se can improve the CD44 expression.
