摘 要: | ![]() 目的检测电离辐射照射HepG2细胞后,其Nucleolin和p53蛋白的变化情况,结果将为进一步揭示电离辐射对p53信号传导通路的影响提供重要的细胞学基础,并为辐射生物效应研究提供新的生物学依据。方法电离辐射照射HepG2细胞后,分别提取总蛋白、胞浆蛋白和胞核蛋白,然后采用流式和WesternBlot的方法进行蛋白检测。结果HepG2细胞接受4Gy电离辐射照射后,其总蛋白中的Nucleolin和p53表达水平增高,且与照射时间呈正相关;HepG2细胞接受电离辐射照射后的2h、4h和8h胞核蛋白中的Nucleolin和p53表达水平下降,而胞浆蛋白中的Nucleoiin和p53表达水平对应增多。结论HepG2细胞受到一定剂量的电离辐射刺激后,Nucleolin和p53在肿瘤细胞内有明显的由胞核至胞浆的穿梭作用,二者的穿梭具有一定的协同性。

Abstract: | ![]() Objective To detect the changes of Nucleolin and p53 proteins in HepG2 cells after irradiation. In order to provide important cytological basis for revealing the effects of radiation on the p53 signaling pathway and provide new biological basis for the study of the radiation effect of cell. Methods HepG2 cells were treated with irradiation, then we extracted the total proteins, cytoplasma, neucleoprotein and detected them by using the method of flow cytome- try and Western Blot. Results The expression level increased of Nucleolin and p53 proteins in total proteins after HepG2 cells were irradiated with 4 Gy, the changes were associated with the irradiation time. The expression level of Nucleolin and p53 in the nuclei of HepG2 cells after 2 h,4 h and 8 h irradiation decreased but correspondingly increased in the cytoplasm. Conclusion There are obvious movements of Nucleolin and p53 from the nucleus to the cytoplasm af- ter the tumor cells wew treated with certain irradiiation and there is cooperativity between them. |
