Study on Changes of Chemical Composition of Alkaloids of Gelsemium elegans Based on Didang Decoction after Reducing Toxicity

Abstract:Objective To establish a method for determination of alkaloids of Gelsemium elegans in vitro and in vivo;and to study the content of alkaloid components of Gelsemium elegans before and after reducing toxicity;To explore the possible mechanism of the action of Didang Decoction in the process of reducing toxicity.Methods UPLC-QDa method was used to detect the changes of 6 alkaloids after combined use of Gelsemium elegans and Didang Decoction in vitro.UPLC-MS/MS method was established to detect the changes of plasma alkaloids in mice fed with Gelsemium elegans combined with Didang Decoction.Results The contents of koumine,gelsemine,gelsevirine and humantenine of Gelsemium elegans were signifantly decreased under the action of Didang Decoction(P<0.01).The contents of koumine,gelsemine,gelsenicine,gelsevirine,humantenine and humantenidine in group of Didang Decoction+Gelsemium elegans were significantly decreased compared with Gelsemium elegans group(P<0.01).Conclusion The decrease of koumine,gelsemine,gelsenicine,gelsevirine and humantenine of Gelsemium elegans may be one of the mechanisms of Didang Decoction to reduce the toxicity of Gelsemium elegans.
