摘    要:目的基于TCGA数据库分析ALAS1在肝癌中的表达及临床意义。方法从TCGA数据库中下载肝癌组织和正常肝组织的RNASeq数据以及临床数据和预后数据,分析ALAS1 mRNA在肝癌组织和正常肝组织中的表达差异以及肝癌中ALAS1基因表达水平与临床病理特征及预后的相关性。LinkedOmics数据库分析与ALAS1表达相关的基因。GSEA软件分析预测ALAS1可能的调控信号通路。结果肝癌组织中ALAS1 mRNA表达水平(12.39±1.20)显著低于正常肝组织(13.93±0.72),差异具有统计学意义(t=8.87,P<0.01)。ALAS1 mRNA的表达水平与性别、T分期以及AJCC分期显著相关(χ2分别为31.39、6.86和6.77,P均<0.01),而与年龄、N分期和M分期无相关。生存分析表明,低表达ALAS1患者的生存期显著低于高表达患者。此外,ALAS1表达与POR、SORD、AQP9基因呈显著正相关(r分别为0.70、0.68和0.67,P均<0.01),而与PKM2、VEGFB、PLCB3基因呈显著负相关(r分别为-0.65、-0.60和-0.60,P均<0.01)。ALAS1的功能分析显示,低表达样本显著富集到细胞周期、T细胞受体信号通路、GnRH信号通路以及趋化因子信号通路等基因集。结论ALAS1基因可能作为肝癌临床预后评估的指标以及靶向治疗的新靶点。

Abstract:Objective To analyze the expression and clinical significance of 5-aminolevulinate synthase 1(ALAS1)in liver cancer based on The Cancer Genome Atlas(TCGA)database.Methods The RNASeq data,clinical data and prognosis data of liver cancer and normal liver tissues were downloaded from TCGA database.The expression difference of ALAS1 mRNA in liver cancer and normal liver tissues,and the correlations of the expression levels of ALAS1 in liver cancer with clinical pathological characteristics and prognosis were analyzed.The genes related to the expression of ALAS1 were analyzed with LinkedOmics database.The possible regulatory signaling pathways of ALAS1 were predicted by the gene set enrichment analysis(GSEA).Results The expression levels of ALAS1 mRNA in liver cancer tissues(12.39±1.20)were significantly lower than that in normal liver tissues(13.93±0.72,t=8.87,P<0.01).The expression levels of ALAS1 mRNA were significantly correlated with gender,T staging and AJCC staging(χ2 values were 31.39,6.86 and 6.77,respectively,all P<0.01),but not with age,N staging and M staging.Survival analysis showed that the survival time of patients with low expression of ALAS1 was significantly lower than that of patients with high expression.In addition,the expression levels of ALAS1 were positively correlated with those of POR,SORD and AQP9(r values were 0.70,0.68 and 0.67,respectively,all P<0.01),and negatively correlated with those of PKM2,VEGFB and PLCB3(r values were-0.65,-0.60 and-0.60,respectively,all P<0.01).The function analysis of ALAS1 showed that low expression samples were significantly enriched in cell cycle,T cell receptor signaling pathway,GnRH signaling pathway and chemokine signaling pathway.Conclusion ALAS1 gene may be a new target for the clinical prognosis and targeted therapy of liver cancer.
