摘    要:目的探讨间皮细胞在腹膜抗菌防御中的作用。方法采用体外细胞培养模型,以间接免疫荧光技术和细胞瑞氏染色方法,观察了细菌脂多糖(LPS)、金葡菌肠毒素和白细胞介素-1(IL-1)诱导大鼠腹膜间皮细胞粘附分子-1(ICAM-1)的表达,及其对金葡菌粘附和单核细胞表面吞噬作用的影响。结果腹膜间皮细胞表达ICAM-1粘附分子,用LPS、金葡菌肠毒素和IL-1刺激培养6小时可增加间皮细胞ICAM-1的表达,同时可增加细菌对间皮细胞的粘附率。单核细胞对粘附间皮细胞表面的调理与未调理细菌均能很好地吞噬,两者吞噬率比较无明显差异(P>0.05)。结论炎症介质和炎性细胞因子通过刺激间皮细胞ICAM-1的表达增加,增强间皮细胞对细菌的粘附,为单核细胞吞噬粘附其表面的细菌提供一个适宜的环境

Abstract:Objective To study the role of the mesothelial cells (MCs)in peritoneal antibacterial defence. Methods The MCs from rat omentum were seperated and cultured in vitro. The expression of the adhesion molecule on the surface of MCs incubated in RPMI 1640 medium alone and stimulated by IL 1, LPS or intestinal toxin was assessed. The effects of IL 1, LPS or intestinal toxin on the rate of S.aureus Cowan I adherence to MCs monolayers as well as the rate of phagocytosis of S.aureus by monocytes were observed. Results MCs incubated in RPMI 1640 medium alone released trace ICAM 1 as measured by indirect immunofluorescence. The ICAM 1 level was significantly increased when MCs was stimulated by IL 1, LPS or intestinal toxin for six hours, so was the adherence rate of S.aureus to MCs monolayers. Adherence of S.aureus to mesothelial monolayers increased significantly when the becteria were opsonised with pooled human serum, but the phygocytosis by monocytes occurred indepandently. Conclusion The MCs express high level of ICAM 1 and increase the adherence of bacteria in early peritoneal inflammation so as to provide a favourable surface for the elimination of bacteria invading peritoneal cavity.
