三氧化二砷对人肝癌细胞生长及其尿型纤溶酶受体表达的体外研究

摘    要:目的:体外研究三氧化二砷(As2O3)对人肝癌细胞株SMMC-7721生长及其表达尿型纤溶酶受体(uPAR)的影响。方法:SMMC-7721在不同浓度的As2O3作用不同时间后,采用MTT法检测细胞增殖能力,流式细胞仪检测细胞周期,实时荧光定量聚合酶链反应(PCR)分析As2O3对uPARmRNA表达的影响,并设立不加As2O3的对照组。结果:As2O3对SMMC-7721细胞的生长呈明显的抑制作用,且呈时间、浓度依赖性;与对照组比较,As2O3实验组G0/G1期细胞比例明显降低,G2/M期细胞比例提高(P<0.05),uPARmRNA表达量明显减少(P<0.01)。结论:As2O3能有效抑制肝癌细胞株SMMC-7721的生长,下调uPARmRNA的表达可能是其作用机制之一。

Effect of Arsenic Trioxide on Human Hepatoma Cell Proliferation and Its uPAR Expression

Abstract:OBJECTIVE: To investigate the effect of arsenic trioxide (AS2O3) on human hepatoma cells SMMC-7721 viability and the expression of urokinase plasminogen activator receptor (uPAR). METHODS: SMMC-7721 cells were cultured and treated with As2O3at different concentrations for different time with untreated SMMC 7721 cells (without AS2O3 treatment) served as control. The proliferation rate of the SMMC-7721 cells was measured by MTT assay; the change of cell cycle was detected by flow cytometry analysis; the effect of AS2O3 on uPAR mRNA levels was analyzed by real - time fluorescent quantitative polymerase chain reaction. RESULTS: AS2O3 had significant inhibitory effect on SMMC - 7721 proliferation in time- dependent and concentration- dependent manner. As compared with control group, AS2O3 significantly decreased the proportion of cells in G0/G1 phase but increased the cell proportion in G2/M phase, and it markedly downregulated the level of uPAR rnRNA expression (P 〈0.01). CONCLUSION: The inhibitory effect of As2O3 on the proliferation of SMMC- 7721 is possibly due to the down - regulation of uPAR mRNA level expression.
