iNOS表达与喉癌组织中淋巴管生成及血管生成的相关性分析

目的:探讨诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)在喉癌中的表达及其与喉癌淋巴管生成、血管生成及预后的相关性。方法:应用免疫组化法和实时荧光定量PCR方法检测iNOS在45例喉癌和20例声带息肉组织中的表达,5’-核苷酸酶染色法计数淋巴管密度(lymphatic microvessel density,LVD),CD34染色计数微血管密度(microvessel density,MVD),并结合临床病理特征和生存资料进行相关分析。结果:喉癌组织中iNOS的表达较声带息肉组显著增加(P<0.05),iNOS表达与瘤内和瘤周LVD、MVD、淋巴结转移、TNM临床分期、淋巴管侵润有关(P<0.05)。iNOS阳性表达与生存率负相关(P<0.05)。结论:喉癌组织中iNOS过表达能够促进喉癌淋巴管生成和血管生成。检测iNOS表达可成为判断喉癌预后新的生物学指标。

Correlation of iNOS and lymphangiogenesis, angiogenesis in laryngeal carcinoma

Objective:To investigate the expression of inducible nitric oxide synthase(iNOS) in patients with laryngeal carcinoma,and the relationship with lymphangiogenesis,angiogenesis and their prognosis significance.Methods: The expressions of iNOS in 45 specimens of the laryngeal cancer and 20 specimens of polyp of vocal cord tissues were observed by immunohistochemistry and real-time fluorescent quantitative PCR.LVD was evaluated by 5′-Nucleotides staining,MVD was evaluated by CD34,and the correlation analysis was made between iNOS and clinical patho-character.The role of iNOS in laryngeal carcinoma prognosis was analyzed by Kaplan-meier.Results: The expression of iNOS was significantly higher in laryngeal cancer tissue than in the polyp of vocal cord tissue(P<0.05),the expression of ET-1 was significantly correlated with intratumoral and peritumoral LVD,MVD,lymph node metastasis,lymphovascular invasion,and TNM clinical stage(P<0.05).The expression of iNOS was correlation with the survival ratio(P<0.05).Conclusions:The expression of iNOS is correlated with lymphangiogenesis and angiogenesis.Detection the expression of iNOS may be helpful for judging prognosis in laryngeal cancer.
