摘    要:目的:探讨小型化的免疫偶联物Fab-ADM的抗肿瘤机制。方法:建立小鼠肝癌细胞H22及小鼠黑色素瘤细胞B16的皮下移植瘤模型,分别予以生理盐水及ADM、Fab-ADM治疗,治疗结束后取肿瘤组织制备石蜡切片进行HE染色,并通过CD34进行免疫组织化学染色以计数肿瘤微血管。结果:在2种动物模型中,Fab-ADM治疗组小鼠瘤体内可见肿瘤细胞的大片状坏死,微血管密度明显减少,与生理盐水对照组及ADM治疗组相比差异有统计学意义(P<0.05)。结论:Fab-ADM偶联物较游离ADM具有更强的肿瘤抑制作用,其可显著抑制肿瘤微血管密度的增加,继而引起肿瘤细胞的大面积坏死。

Abstract:Objective: To investigate the anti-tumor mechanism of dowmsized immunoconjugate Fab-ADM.Methods: Preparing hepatocarcinoma H22 and melanoma B16 tumor-bearing mice and separated them into three groups,treated with normal saline,adriamycin and immunoconjugate Fab-ADM,respectively.SP immunohistochemistry method was used to detect the expression of CD34.Results: For two mice tumor models,the tumor cells appeared large area necrosis in Fab-ADM treated group,and microvessel density also decreased significantly(P<0.05) compared with the normal saline and adriamycin treated groups.Conclusion: Treatment with Fab-ADM supressed increasing of tumor microvessel density significantly,and then causeed large area necrosis,which may be the mechanism of Fab-ADM treatment effection.
