摘    要:采用超高效液相色谱建立三叶青叶指纹图谱,并同时测定8种酚类成分(新绿原酸、绿原酸、隐绿原酸、异荭草苷、荭草苷、牡荆素鼠李糖苷、牡荆苷、异牡荆苷)的含量,为三叶青叶质量评价提供依据。采用UHPLC法,Welch UHPLC Ultimate XB-C18色谱柱(4.6 mm×150 mm,2.7μm),流动相为乙腈-0.1%甲酸(含5%甲醇)水梯度洗脱,流速0.4 m L·min~(-1),柱温30℃,采用国家药典委员会"相似度评价软件2004AB"处理分析,建立41批不同产地三叶青叶的指纹图谱,共标定15个共有峰并对不同产地进行聚类分析,然后选择其中8个主要的酚类成分作为定量指标,建立UHPLC同时测定8种主要酚类成分含量的方法,经方法学验证该研究建立的三叶青叶指纹图谱和8种酚类成分含量测定方法简便有效、灵敏、准确,可为综合评价三叶青叶的质量提供参考。

Abstract:A novel method combining ultra-high performance liquid chromatography(UHPLC) fingerprint and simultaneous quantitative analysis of eight phenolic components was developed and validated for quality evaluation of Tetrastigma hemsleyanum leaves. For fingerprint analysis, 15 peaks were selected as the common peaks to evaluate the similarities among 41 batches of T. hemsleyanum leaves collected from different regions. Additionally, simultaneous quantification of eight markers, including neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isoorientin, orientin, vitexin-2-O-rhamnoside,vitexin and isovitexin, was performed and the obtained data demonstrated that our method has achieved desired linearity, precision and accuracy. Clustering statistical analysis was further application in T. hemsleyanum leaves from different regions. The results indicated that new approach conbine ultra-high performance liquid chromatography(UHPLC) fingerprint and simultaneous quantitative analysis of eight phenolic components was applicable in quality control of T. hemsleyanum leaves.
