冬虫夏草中游离麦角甾醇的研究

摘    要:目的:建立冬虫夏草药材中麦角甾醇的RP—HPLC含量测定方法。方法:冬虫夏草药材粉末经甲醇超声提取后,用RP—HPLC测定。采用Phenomenex Luna C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-水(98:2);流速为1mL·min^-1;检测波长为282nm。结果:麦角甾醇进样量在0.04272~0.4272μg与峰面积呈良好的线性关系(r=0.9999);麦角甾醇的平均回收率为98.7%(n=6)。结论:采用建立的RP-HPLC方法测定冬虫夏草药材中麦角甾醇的含量,方法灵敏、快速、准确。

Abstract:Objective: To establish a RP-HPLC method for quantitative analysis of ergosterol in Cordyceps. Methods:A Phenomenex Luna C18(250 mm ×4. 6 mm,5 μm) column was used with the mobile phase being methanol-water ( 98 : 2 ), flow rate being 1.0 mL·min ^- 1, detecting wavelength being 282 nm . Results: The method had good linear relationship within the range of 0. 042 72 - 0. 427 2 μg ( r = 0. 999 9 ). The average recovery was 98.7% (n = 6),RSD =4. 5%. Conclusion:The method is sensitive,quick and accuracy.
