姜黄及其3种近源品种的姜黄素类成分薄层色谱鉴别及HPLC特征图谱分析

摘    要:目的:鉴别分析姜黄及其3种近源品种(蓬莪术、温莪术、广西莪术)的姜黄素类成分。方法:以姜黄对照药材,姜黄素、去甲氧基姜黄素、双去甲氧基姜黄素对照品为对照进行薄层色谱鉴别;以姜黄素、去甲氧基姜黄素、双去甲氧基姜黄素对照品为对照物质,采用HPLC对10批姜黄进行特征图谱研究,建立HPLC特征图谱共有模式,进行相似度比较,并将姜黄与3种近源品种进行相似度比对。色谱条件:采用月旭AQ-C18(200mm×4.6mm,5μm)色谱柱,以乙腈-水为流动相进行梯度洗脱,流速1mL·min^-1,检测波长270nm,柱温30℃。结果:供试品色谱中,姜黄与3种莪术的斑点位置、颜色、数量有明显差别;建立了姜黄的HPLC特征图谱,确定了7个共有峰,10批姜黄药材样品的平均相似度在0.97,姜黄与3种近源品种的相似度均低于0.8,特征图谱差异明显。结论:姜黄及其3种近源品种的薄层色谱及HPLC特征图谱差异明显,且方法简便、准确、专属性强,可作为姜黄及其3种近源品种的鉴别,同时也为姜黄对照药材标定技术标准的建立提供了科学依据。

Abstract:Objective:Researching Curcumae longae rhizoma and its three near-source species and ldentificate the kinds of the curcuminoids, netherds: TLC was used to identificate curcuminoids in Curcumae longae rhizoma and its three near-source species, with Curcumae longae rhizoma, Curcumine, Demethoxycurcumin, Diplo-demethoxycurcumin as reference substance. We established a HPLC fingerprint chromatogram to identificate the difference from near-source species. Results:The distinction of Curcumae longae rhizoma, and its three near-source species are obviously difference and can be identificated by TLC and HPLC fingerprint chromatogram. Conclusion: The identification methods were proved to be available and effective.
