保达新对缺血再灌注损伤大鼠心肌的影响

摘    要:目的 观察保达新对缺血再灌注损伤心肌的保护作用.方法 将24只左冠状动脉前降支(LAD)结扎大鼠分成3组(假手术组、对照组、治疗组),分别在心肌缺血30 min后再灌注60min,复灌前5 min静脉注射0. 9%生理盐水0.25 ml/kg;或保达新0 .5 μg/kg,检测血液中乳酸脱氢酶(LDH)、血浆丙二醛(MDA)、白细胞介素(IL)-6及IL-8的差异和各组间bcl-2、bax蛋白表达的差异.结果 对照组与其他两组比较LDH、MDA、IL-6及IL-8明显增加(P<0.05),治疗组中LDH、MDA、IL-6及IL-8较空白组明显增加(P<0.05).对照组与假手术组比较bcl-2、bax蛋白表达明显增加(P<0.05),治疗组中bcl-2表达较其他两组明显增加(P<0.05),bax表达较对照组明显减少(P<0.05).结论 保达新可通过减少脂质过氧化物产生、减轻过氧化反应,抑制细胞凋亡来保护缺血再灌注损伤的心肌.

Abstract:Objective To investigate the protective effect of Prostavasin on Myocardial ischemia-reperfusion injury. Methods 24 left anterior descending coronary artery (LAD) ligated rats were divided into 3 groups (sham operation group,control group,treatment group). Reperfused 60 min respectively after myocardial ischemia 30 min,5 min before reperfusion intravenous 0.9% saline 0.25 ml/kg or Prostavasin 0.5μxg/kg. Detected the differences between blood Iactate dehydrogenase (LDH), plasma malondialdehyde (MDA) ,interleukin (IL)-6 and IL-8,and the differences of bcl-2,bax protein expression between each group. Results Compared with other two groups, LDH, MDA, IL-6 and IL-8 increased significantly in the control group (P<0.05) ;LDH,MDA,IL-6 and IL-8 in treatment group more significantly increased than the blank group(P<0.05). Compared with the sham operation group protein expression of bcl-2 and bax were significantly increased in control group (P < 0.05); in treatment group, the expression of bcl-2 significantly increased compared with the other two groups, bax expression were markedly reduced (P < 0.05). Conclusion Prostavasin can protect the ischemic-reperfusion injury myocardium by reducing lipid peroxides and reducing peroxidatic reaction, inducing apoptosis.
