紫杉醇对血管外膜成纤维细胞增殖和迁移的影响

摘    要:目的观察紫杉醇对血管外膜成纤维细胞(AF)增殖、表型转化和迁移的影响。方法选取雄性SD大鼠,用贴壁法培养胸主动脉外膜AF,用免疫荧光法检测AF增殖细胞核抗原(PCNA),用流式细胞仪检测细胞周期,用Transwell法测定细胞迁移能力。结果 10 nmol.L-1紫杉醇对成纤维细胞存活率较空白组无明显差异,表明其对成纤维细胞无明显杀伤作用。与空白组比较,9 nmol.L-1紫杉醇可显著增加G0/G1期AF细胞百分比(P<0.05),降低G2/M及S期细胞百分比(P<0.05),并显著降低AF细胞PCNA表达,抑制其迁移能力(P<0.01)。结论低浓度紫杉醇在不影响成纤维细胞生存活力的情况下,能显著抑制细胞增殖和迁移能力。

Abstract:Objective To study on the migration and proliferation of vascular adventitial fibroblasts.Methods Adventitial fibroblasts(AF) were isolated from thoracic aortas of male Sprague-Dawley rats.The proliferation of adventitial fibroblasts was examined on the proliferating cell nuclear antigen(PCNA) by immunofluorescence staining.Analysis of the cell cycle is performed by flow cytometry.The ability of cell migration was detected by chamber invasive models.Results Low dose paclitaxel(≤10 nmol·L-1) for 12 hours have no significant decrease in AF survival rate when compared with control group(P<0.05).The percentage of G0-G1 phase cells increased while the G2-M and S phase decreased after incubation with paclitaxel(9 nmol·L-1) for 24 hours(P <0.05).Paclitaxel significantly inhibit the PCNA expression as well as the migration ability in AF(P<0.01).Conclusion Low dose of paclitaxel evidently inhibit the proliferation and migration of AF,therefore inhibit restenosis after coronary stenting.
