环氧化酶2在鼻咽癌中的表达及其与血管生成和细胞增殖的关系

摘    要:
目的 探讨环氧化酶2(COX-2)、血管内皮生长因子(VEGF)在鼻咽癌中的表达及其与血管生成和细胞增殖的关系.方法 标本取自我科收治的62例鼻咽癌患者,应用免疫组织化学方法(SABC法)检测COX-2和VEGF的表达,并检测微血管密度(MVD)和增殖细胞核抗原(PCNA)指数.结果 COX-2和VEGF在鼻咽癌中的表达率分别为77.42%(48/62)和53.23%(33/62),主要在癌组织中表达,而在癌旁组织中少见表达;有淋巴结转移组中,COX-2和VEGF的表达水平均高于无转移组(P<0.01).COX-2表达阳性者的MVD和PCNA指数明显高于COX-2表达阴性者(P<0.05).结论 鼻咽癌中COX-2和VEGF的高表达与鼻咽癌的发生、发展及淋巴结转移有重要关系.

Abstract:
Objective To observe the expressions of cyclooxygenase 2 (COX-2) and vascular endothelial growth factor(VEGF) and their relationships with neovascularization and tumor cell proliferation in nasopharyngeal carcinoma. Methods The expressions of COX-2, VEGF, microvascular density (MVD) and proliferating cell nuclear antigen (PCNA) were examined by immunohistochemical technique( SABC method)in 62 eases with nasopharyngeal carcinoma. Results The positive rates of COX-2 and VEGF in nasopharyngeal carcinoma was 77.42% and 53.23% respectively. But there was little expression of them in noncancerous specimens of nasopharyugus. The expression rates of COX-2 and VEGF were higher in the group of lymph node metastasis than those in the group of without lymph nodes metastasis(P 〈 0.01 ). The MVD and PCNA indexes were significantly higher in the COX-2 positive tumors than in the negative ones (P 〈 0.05 ). Conclusions The over-expressions of COX-2 and VEGF might play a crucial role in the carcinogenesis and development of nasopharyngeal carcinoma, which might enhance lymphatic metastasis in patients with nasopharyngeal carcinoma.
