镍钛合金裸支架对内皮细胞低密度脂蛋白穿胞影响的初步研究

摘    要:
目的探讨镍钛合金裸支架是否影响低密度脂蛋白(LDL)在人脐静脉内皮细胞(HUVEC)中的穿胞并初步探讨可能的机制。 方法①培养HUVEC,利用细胞培养小池模拟血管内皮单层细胞,建立穿胞模型;②将镍钛合金裸支架作用于HUVEC上,加入异硫氰酸荧光素(FITC)标记的LDL,分别培养3 h及24 h;③通过测定培养小池内外FITC-LDL的量,研究镍钛合金裸支架对LDL在HUVEC中穿胞过程的影响。 结果3 h及24 h时镍钛合金裸支架作用下LDL在HUVEC中的穿胞量均增加。 结论在LDL和HUVEC构建的穿胞模型实验中,镍钛合金裸支架可以促进LDL的穿胞。

Abstract:
ObjectiveTo explore the effects of Ni-Ti alloy stent on the low density lipoprotein (LDL) transcytosis process in human umbilical vein endothelial cell (HUVEC) and underlying mechanism. Methods①Culturing the HUVEC and use transwell to establish in vitro transcytosis model, simulating the endothelial cell monolayer of blood vessels. ②Puting the Ni-Ti alloy stent on the HUVEC, adding LDL labeled by fluorescein isothiocyanate (FITC), culturing for 4 h and 24 h. ③By measuring the fluorescencent intensity of FITC in and out of the chamber to reflect the changes of transcytosis of LDL, investigating the effect of the Ni-Ti alloy stent on the LDL transcytosis process. ResultsThe Ni-Ti alloy stent increase the transcytosis in 3 h and 24 h. ConclusionsUsing the trancytosis model constituted by LDL and HUVEC, Ni-Ti alloy stent can promote the trancytosis of LDL.
