七叶皂苷钠对大鼠局灶性脑缺血损伤炎症反应的作用

摘    要:目的:观察七叶皂苷钠对大鼠脑缺血再灌注损伤局部炎症反应的影响。方法:线栓法阻塞大鼠左侧大脑中动脉,建立缺血再灌注模型,经视交叉的冠状脑组织块行TTC染色测梗死面积,并进行髓过氧化物酶(myeloperoxidase,MPO)、细胞间黏附分子-1(inter-cellular adhesion molecule-1,ICAM-1)免疫组化染色。结果:再灌注即刻腹腔内注射七叶皂苷钠的治疗组较对照组神经功能缺损程度显著减轻(P<0.05),梗死面积明显缩小(P<0.05),MPO阳性细胞数明显减少(P<0.05),ICAM-1免疫阳性血管数无显著变化(P>0.05)。结论:七叶皂苷钠对局灶性脑缺血损伤的保护作用可能与抑制局部炎性渗出有一定的关系。

Abstract:Objective:To investigate the effect of sodium aescinate on inflammation in focal cerebral ischemia rats. Methods:Male SD rats were subjected to middle cerebral artery occlusion for 2 h and reperfusion for 22 h with an intraluminal vascular occlusion method. TTC staining was used to calculate the infarct size at the optic chiasma level. Myeloperoxidase (MPO) and inter cellular adhesion molecule 1(ICAM 1) immunohistochemical stain were also used to detect the changes of neutrophil and vessel density. Results:In contrast to control rats, rats treated with sodium aescinate had smaller infarct areas, lower neurological deficit scores and fewer MPO positive cells( P <0.05), while the number of ICAM 1 positive vessels had no significant difference( P >0.05). Conclusion: These data suggest that sodium aescinate attenuate focal ischemic damage, and the positive effect may be partly due to its anti inflammatory property.
