HPC和L929细胞在修复粘结剂体外毒性评价中灵敏度的比较研究

摘    要:目的:通过原代培养的人牙髓细胞(HPC)和L929细胞系,对两种树脂改良型玻璃离子粘结剂的体外细胞毒性评价,比较两种细胞的灵敏度,为口腔材料的体外评价建立更接近临床的评价体系。方法:制取Fujiplus和RelyX Luting两种材料浸渍液,实验组为材料的浸出原液、50%原液、25%原液,对照组为DMEM培养基,分别利用原代培养的牙髓细胞和L929细胞系对实验组和对照组进行细胞形态观察、MTT测定细胞相对增值率。结果:树脂改良型玻璃离子粘结剂的体外细胞毒性评价中,和选择的细胞有关,原代培养的牙髓细胞较L929细胞系敏感。结论:原代培养的人牙髓细胞可能较L929细胞系更适用于口腔粘材料的体外毒性评价。

Abstract:Objective: The aim of this study was to compare the sensitivity by two resin-modified glass-ionomer cements(RMGIC) toward primary dental human pulp cells(PHPCs) and L929 cell lines in vitro.Methods: The two kinds of RMGICs enrolled were Fuji plus and RelyX luting.Elutes were prepared by placing specimens in Petri dishes with DMEM cell culture medium.PHPCs and L929 cells were seeded into 96 well culture plates.The extracts in 100%,50% and 25% dilution were used as test groups.The fresh DMEM was used as control group.Morphology of cells was examined by an inverted microscope.Cell numbers were measured with a modified 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assay.Results: According to the methodology employed in the present study,the cytotoxicity of RMGICS in vitro varied based on the materials tested and on the cell lines selected.primary human pulp cells show more sensitivities to the elutes of RMGICs and RelyX luting cement.PHPCs cells were more sensitive to the cement extracts than L929 cells.Conclusion: primary human pulp cells may be more suitable than the L929 cells for the cytotoxicity of resin-modified glass-ionomer cements in vitro.
