FAF1在胃癌组织中的表达及其临床意义

目的：研究FAF1在胃癌组织中的表达及其临床意义。方法：分别采用荧光实时定量PCR（FQ-RT-PCR）和免疫组织化学方法检测40例胃癌组织、癌旁组织及相对正常胃黏膜组织FAF1mRNA和FAF1蛋白的表达。结果：FAF1mRNA在胃癌组织（O．27±0．12）中的表达水平明显低于癌旁（0．40±0．06）和正常胃组织（0．48±0．08）（P〈0．01）。FAF1mRNA在胃癌中的表达水平与癌细胞分化程度、远处转移有关,分化程度为低未分化及远处转移的胃癌组织FAF1mRNA表达值明显低于高中分化及无远处转移者,而与年龄、性别、浸润深度、临床分期、有无淋巴结转移及肿瘤大小无关。胃癌组织的FAF1蛋白的阳性表达率（37．50%）明显低于癌旁组织（62．50％）（P〈O．05）和正常组织（72．50％）（P〈0．01）;FAF1蛋白在胃癌中的表达水平与癌细胞分化程度有关,低或未分化胃癌组织FAF1蛋白阳性表达率显著低于高中分化胃癌组织,而与年龄、性别、肿瘤大小、浸润深度、淋巴结转移、远处转移和临床分期无关。结论：胃癌组织中存在FAF1表达减少,可能与胃癌的发生、发展有关。FAF1可以作为一个判断胃癌预后的指标,也可能成为胃癌基因治疗的一个新靶点。

EXPRESSION OF FAF1 IN GASTRIC CANCER TISSUE AND ITS CLINICAL SIGNIFICANCE

Objective： To explore the relationship between the expression of FAF1 in gastric cancer tissue and its clinical significance. Methods：The mRNA and protein expression of FAF1 were detected by fluorescent quantitative real time polymerase chain reaction（FQ-RT-PCR） and immunohistochemistry in 40 cases of gastric cancer, paracancerous, and normal tissues ; respectively. Results ： The expression level of FAF1 mRNA in the cancerous tissues （0.27±0. 12） was markedly lower than that in the paracancerous tissues（0.40±0.06） and normal tissues （0.48±0.08）（ P 〈0.01）. The expression of FAFlmRNA in gastric cancer tissue was closely correlated with cell differentiation and distantmetastasis. The level of FAFlmRNA was lower in poorly-differentiated and distantmetastasis cancerous tissues than that in well-differentiated and no distantmetastasis ones,but it was not correlated with age, sex, depth of invasion, stage （TNM）, lymph node metastasis and the size of tumor. The expression level of FAF1 protein was significantly lower in the gastric cancer tissues （37.50%） than that in the para-cancerous tissue （62.50%, P 〈0.05） and normal tissue （72.50%, P 〈0.05）. The expression of FAF1 protein was lower in poorly-differentiated cancerous tis sues than that in well-differentiated ones （ P〈0.05） ,but no correlation with age, sex, depth of invasion, stage （TNM）,lymph node metastasis,the size of tumor and distantmetastasis was found. Conclusion：Decreased expression of FAF1 may play an important role in the occurrence and development of gastric cancer. FAF1 may be an index of prognosis,and also a new target of gene therapy in gastric cancer.
